摘  要:目的:观察黄连总生物碱对糖尿病大鼠血管内皮细胞的影响。方法:链脲佐菌素一次性腹腔注射复制糖尿病大鼠模型。给药组灌胃给予黄连总生物碱(50,100,200mg/kg)、格列齐特(20mg/kg),正常组和模型组给予等容量生理盐水,每天1次,连续8周。实验结束后,分光光度法检测大鼠血糖水平,放射免疫法测定6-酮-前列腺素(6-Keto-PG)F1α、血栓素(TX)B2、血管紧张素(Ang)Ⅱ的含量;ELISA法检测血管性血友病因子(vWF)、纤溶酶原激活物抑制剂(PAI)-1的水平;取胸主动环观察不同累积浓度的乙酰胆碱(Ach)对去甲肾上腺素(NE)引起的血管收缩的抑制率;免疫组化技术测定主动脉细胞间粘附分子(ICAM)-1蛋白的表达。结果:黄连总生物碱不仅能显著降低糖尿病大鼠血糖水平、血浆TXB2、AngⅡ、vWF、PAI-1含量,升高6-Keto-PGF1α的水平,改善糖尿病大鼠内皮依赖性舒张功能,同时还能有效抑制糖尿病大鼠主动脉ICAM-1蛋白的表达。结论:黄连总生物碱对糖尿病大鼠血管内皮细胞具有一定的保护作用。Objective: To observe the effect of vascular endothelium cells by extraction total alkaloids from Coptidis Rhizome(TCA) in diabetic rats.Methods: Experimental diabetic rats was induced by streptozotocin.All the therapeutic groups were treated with TCA(50,100,200 mg/kg) and the gliclazide(20mg/kg) respectively for 8 weeks.The blood sugar level were determinate by chromatometry.The 6-keto-prostaglandin(6-Keto-PG)F1α,thromboxan(TX)B2,angiotonin(Ang)Ⅱcontent were assayed by radioimmunity,thevon Willebrand disease factor(vWF),plasminogen activator inhibitor(PAI)-1 level were determinate by enzyme linked immunosorbent assay.Thorac-ic aortic ring was employed to observe the inhibition by different concentration of acetylcholine(Ach) to constraction caused by norepinephrine(NE),SP immunohistochemicae staining was employed to observe the expression of intercellular adhesion molecule(ICAM-1).Results: TCA not only could decreased the blood sugar,TXB2,AngⅡ,vWF,PAI-1content,reduced the inhibition of vasodilator responses,step up 6-Keto-PGF1α activities,but also effectively prevented the expression of ICAM-1.Conclusion: TCA has a protect function on injury of vascular endothelium cells in diabetic rats.
