狭基线纹香茶菜对免疫性肝损伤小鼠TNF-α mRNA和蛋白表达水平的影响

摘　　要：目的:以Con A诱导小鼠免疫性肝损伤模型,研究狭基线纹香茶菜对免疫肝损伤小鼠TNF-αmRNA和蛋白表达的影响,以探讨其抗免疫肝损伤的作用机理。方法:小鼠尾静脉注射ConA 25mg/kg制备免疫肝损伤模型,灌胃给予狭基线纹香茶菜水提部位7d后,不同时间点采取肝组织,提取总RNA,用反转录聚合酶链式反应(RT-PCR)对肝脏TNF-αmRNA进行半定量测定,免疫组化法检测肝组织TNF-α蛋白的表达。结果:与正常组相比,模型组小鼠肝组织4h TNF-αmRNA和蛋白表达显著增强,至8h仍保持较高的表达。阳性药PDTC和狭基线纹香茶菜水提部位大剂量组TNF-αmRNA和蛋白表达显著低于模型组(起效剂量9.2g/kg)。结论:ConA诱导损伤后,肝组织中TNF-αmRNA和蛋白表达明显增强,狭基线纹香茶菜组可通过降低损伤肝组织TNF-αmRNA和蛋白的表达,而对免疫肝损伤小鼠起保护作用。Objective： Established immune liver injury model in mice induced by ConA to observe influence of water-soluble extract from Isodon lophanthoides var.gerardianus on TNF-αmRNA and protein expression,so that to explore its mechanism of protective action against liver injury.Methods ：Mice were challenged by a single tail intravenous injection of Con A（25mg/kg） to induce immnuce liver injury,meanwhile,water-soluble extract from Isodon lophanthoides var.gerardianus is administrated to mice for 7days,we collected liver tissue in different time point.TNF-αmRNA and protein expression in liver tissue were detected by RT-PCR and immunohistochemistry examination.Results： Compared with normal control group,ConA administration can cause increased hepatic TNF-αeither mRNA or protein expression at 4h,still keep higher level at 8h.The positive group and extract group（9.2g/kg）could significantly lower the protein expression of TNF-α,furthermore,the mRNA expression of in liver tissue were lower than those of model group.Conclusion： Extract from Isodon lophanthoides var.gerardianus may inhibit TNF-αmRNA and protein expression,it might be related to mechanism of protecting action against liver injuty.
