摘 要:目的:基于代谢物组学理论,探讨黄连解毒汤的拆方黄连-黄芩及黄连-黄芩-黄柏干预2,4-二硝基苯酚诱导热病证候模型在代谢物组学层面上的关联性。方法:运用UPLC/MS技术手段,采用PCA和PLS-DA数据解析方法,研究黄连-黄芩及黄连-黄芩-黄柏对于热病证候模型大鼠尿液中生物标记物的影响。结果:初步确定2,4-二硝基苯酚诱导热病证候模型51个生物标志物,黄连-黄芩对于其中8个生物标志物具有明显干预作用,黄连-黄芩-黄柏对于其中15个生物标志物有明显干预作用。结论:黄连-黄芩-黄柏对于热病证候模型生物标志物的干预数量和程度均好于黄连-黄芩,佐药黄柏的加入有效加强了对热病的治疗作用,从代谢物组学的角度阐明了传统复方理论中"君臣佐使"组方原则的科学性。Objective: To explore the effect of decomposed recipes in Huanglianjiedu Decoction on the 2,4-dinitrophenol-induced fever syndrome model based on metabolomics Method:The metabolites in rats urine of 2,4-dinitrophenol-induced fever syndrome model were determined by UPLC/MS and then analyzed by PCA and PLS-DA, comforming the impact of Coptis chinensis - ScuteUaria baicalensis and Coptis chinensis - Scutellaria baicalensis - Cortex Phellodendri . Results: Confirming 51 biomarkers on the 2,4-dinitrophenol-induced fever syndrome model ;Coptis chinensis - Scutellaria baicalensis have effected on 8 biomarkers and Coptis chinensis - ScuteUaria baicalensis - Cortex Phellodendri have effected on 15 biomarkers. Conclusion :The curative effection of Coptis chinensis - Scutellaria baicalensis - Cortex PheUodendri for fever syndrome model is better than Coptis chinensis - Scutellaria baicalensis on quantity and degree of biomarkers. The addition of effective adju- vant Cortex Phellodeudri enhanced the treatment of fever symptoms, and inustrated the scientific of the rules of composing principles of "mon- arch, minister, assistant and guide" from the perspective of metabolomics.
