尿毒净液对慢性肾功能衰竭大鼠肾功能及肾组织中Nephrin、MCP-1和PAI-1表达的影响

摘　　要：目的:观察尿毒净液(Urea Nitrogen Cleaning Fluid,UNCF)对慢性肾功能衰竭大鼠模型肾脏功能的作用及其对肾组织肾病蛋白(nephrin)、单核细胞趋化蛋白-1(MCP-1)和纤溶酶原激活物抑制剂-1(PAI-1)表达的影响。方法:应用5/6肾切除方法复制氮质血症大鼠模型,随机分为模型组、尿毒净液组,以假手术大鼠作为对照组,给药8周后检测各组大鼠血清尿素氮及24小时尿蛋白总量的水平,HE染色法观察肾小球组织形态学改变,免疫组化法、Western blot法及荧光定量PCR法检测各组大鼠肾组织中Nephrin、MCP-1和PAI-1蛋白及mRNA的分布表达。结果:尿毒净液16g/kg连续给药8周显著降低血BUN和尿蛋白,大鼠肾组织中PAI-1、MCP-1表达均显著低于模型组,Nephrin明显高于模型组。结论:尿毒净液能显著改善5/6肾切除大鼠的肾功能,延缓5/6肾切除大鼠慢性肾衰竭的进程,其机理与尿毒净液能抑制MCP-1和PAI-1的表达,同时又促进了Nephrin的表达相关。Objective： To observe the effects of Urea Nitrogen Cleaning Fluid（UNCF） on renal function and expression of nephrin,monocyte chemoatracttant protein-1（MCP-1） and plasminogen activator inhibitor-1（PAI-1） in renal tissues of chronic renal failure（CRF） rats.Methods： 5/6 nephrectomy was used to establish CRF model in rats.UNCF was administrated intragastrically for 8weeks.Blood urea nitrogen（BUN） and 24-hour urine protein was examined.Histomorphology of glomcrulus was observed by H＆E staining method.The expression levels of Nephrin,MCP-1 and PAI-1 in renal tissue were examined in all groups by immunohistochemistry,western blot and fluorescence quantitative PCR.Results ：UNCF lowered the serum BUN and urine protein level significantly.Expressions of PAI-1and MCP-1 in the treatment group were significantly lower than those of the model group while expression of Nephrinin was significant higher than those of model group.Conclusion ： UNCF can improve the kidney function and ameliorate the progression of CRF in rats with 5/6 nephrectomy.These effects are associated with down-regulation of MCP-1 and PAI-1 and upregulation of nephrin in renal tissues.
