摘  要:目的:探讨神经营养因子3(NT-3)在不同发育时期人胚胎脊髓中的表达变化及意义。方法:采用免疫组织化学和免疫印迹法对4周~7月人胚胎脊髓发育过程中NT-3的定位及定量进行研究。结果:NT-3在所检测的11个时段中均有表达,从第6周开始逐渐增加,至第9周时达高峰,之后下降并维持在一定水平,且以细胞突起和纤维表达最明显。结论:NT-3广泛分布于人胚胎脊髓不同发育时期的各种组织中,对胚胎期脊髓的发育起重要作用,并可能对脊髓神经干细胞的分裂、增殖及分化有积极的促进作用。Objective: To explore the expression change and its significance of neurotrophin-3 (NT-3) at different developmental stages of human embryonic spinal cord. Methods: Immunohistochemistry and Western blot were used to systematically study the localization and quantitation of NT-3 in the human embryonic spinal cord from the 4th week to the 7th month. Results: NT-3 positive protein was found in all the detected 11 time points, and concentrated on cell process and fiber. Along with the embryonal age the quantitation of NT-3 increased gradually from the 6th week, reached the peak in the 9th week, and then decreased and maintained at a certain level. Conclusion: NT-3 is widely distributed in human fetal spinal cord of different developmental stages and may play an vital role in the embryonic stage. NT-3 possibly has an positive role in the division, proliferation and differentiation of the spinal neural stem cells.
