摘  要:目的:观察督脉电针联合神经干细胞(NSCs)移植治疗脊髓损伤对硫酸软骨素蛋白多糖aggrecan的表达影响。方法:成年SD大鼠随机分为脊髓损伤对照组、督脉电针组、NSCs移植组以及督脉电针联合NSCs移植组。建立成年大鼠第9~10胸椎段脊髓全横断模型。其中电针组和电针联合NSCs组于术后当天进行电针治疗。用免疫组织化学ABC法和图像分析技术检测脊髓内aggrecan的表达。结果:脊髓损伤对照组aggrecan的表达较正常组增加;术后14d,督脉电针组、NSCs移植组和电针联合NSCs移植组的aggrecan表达较损伤对照组均有减少;术后28d,这3组的aggrecan表达仍保持较低水平;aggrecan的表达变化以督脉电针组和电针联合NSCs移植组最为明显。结论:督脉电针与NSCs移植联合应用能降低脊髓损伤后aggrecan的表达,从而减弱aggrecan对轴突再生的抑制作用,这可能是两者联合治疗促进脊髓损伤修复的可能机制之一。Objective: To explore the effects of combination of goremor vessel electroaeupuncture and transplanted neural stem cells (NSCs) on the expression of aggrecan of chondroitin sulfate proteoglycans in the injured tissues of rat spinal cord transected completely. Methods.. Adult rats were divided into control group, goremor vessel electroacupuncture group (EC group), neural setm cells transplanted group (NSCs group) and goremor vessel electroacupuncture plus NSCs transplanted group (EC+NSCs group). All animals were performed complete transection between T9 and T10 segment of the spinal cord. The electroacupunctural treatment started at the postoperative day among EC group and EC+NSCs group. Aggrecan immunohistochemical staining was made in the tissues of the injured spinal cord. The immuno-positive gray scale of immunoreactivity was detected with computer image analysis technique. Results: The expression of aggrecan at cranial segment and caudal segment of injured spinal cord segment was increased more slightly in the control group than in the normal group. Fourteen days after operation, the expression of aggrecan at the cranial segment and caudal segment of the injured spinal cord segment was decreased more slightly in EC group, NSCs group and EC+NSCs group than in the control group. Twenty-eight clays after operation, the expression of aggrecan at the injured segment maintainted a lower level in EC group, NSCs group and EC+NSCs group; The expression of aggrecan was more prominent in EC group and EC+NSCs group. Conclusion: The combination of goremor vessel electroacupuncture and transplanted neural stem cells can significantly reduce the expression of aggrecan, which is an inhibitor of axon regeneration after spinal cord injury (SCI). This effect may he one of the mechanism that the combination therapy of both can promote the functional recovery after SCI.
