摘 要:目的探讨吸毒合并HIV感染人群与HLA-DRB1基因多态性的相关性。方法应用序列特异性引物聚合酶链反应(PCR-SSP)方法,对无亲缘关系的41例吸毒合并HIV感染者、24例单纯吸毒者及213例健康者进行HLA-DRB1基因型检测,分析正常对照组和吸毒组及吸毒合并HIV感染组HLA-DRB1等位基因频率分布。结果与正常对照组比较,吸毒组中,HLA-DRB1*0401-0411(P<0.05,RR=9.22),*1001(P<0.05,RR=19.27)的等位基因频率明显增高;在吸毒合并HIV感染组中,*0101-0103(P<0.01,RR=7.26)、*0401-0411(P<0.01,RR=16.73)、*0801-0806(P<0.01,RR=5.49)、*1001(P<0.05,RR=22.91)、*1401/1404/1405(P<0.05,RR=5.41)的等位基因频率显著增高,而DRB1*1501-1502(P<0.01,RR=0.18)、DRB5(P<0.01,RR=0.40)的等位基因频率较正常对照组明显减少。与吸毒组比较,吸毒合并HIV感染组中各等位基因频率变化无统计学差异。结论 HLA-DRB1等位基因多态性与吸毒合并HIV感染具有密切相关性,这对阐明吸毒合并HIV感染的发生机制及后期治疗和免疫预防具有重要意义。Objective To investigate the relationship between genetic polymorphism of HLA-DRB1 and Drug taking combined with HIV infection.Methods A total of 41 drug users with HIV infection,and 24 drug users without HIV infection,and 213 healthy controls,all without blood relationship,were selected and analyzed by using polymerase chain reaction with sequence-specific primers(PCR-SSP) to test their HLA-DRB1 allele gene frequencies(GF) and analyze different distribution of HLA-DRB1 alleles.Results The allele GF of the following types were significantly higher than corresponding control:DRB10401-0411(P0.05,RR=9.22),1001(P0.05,RR=19.27)in drug user group;and DRB10101-0103(P0.01,RR=7.26),0401-0411(P0.01,RR=16.73),0801-0806(P0.01,RR=5.49),1001(P0.05,RR=22.91),1401/1404/1405(P0.05,RR=5.41)allele in drug user with HIV infecion group,whereas,the GF of DRB1 1501-1502(P0.01,RR=0.18),DRB5(P0.01,RR= 0.40) in drug user with HIV infection group were significantly lower than in control group.The GF of DRB1 was not statistically differont between drug users with HIV infection and drug users alone.Conclusion There is close relationship between the polymorphism of HLA-DRB1 Alleles and drug taking with HIV Infection,which may play an important role in interpreting the mechanism,and providing the basis for therapeutics and prophylaxis of drug taking with HIV infection.
