刺芒柄花素对去势大鼠主动脉弓雌激素受体β表达的影响

摘　　要：目的:研究植物雌激素刺芒柄花素对去卵巢+高脂血症大鼠主动脉弓雌激素受体β(ERβ)表达的影响以了解刺芒柄花素对去势大鼠心血管的保护作用。方法:实验选取SD大鼠制作去势高脂模型,以Western blot和real-timePCR方法检测主动脉弓中ERβ的含量。结果:Western blot结果表明模型组大鼠主动脉弓ERβ的含量较假手术组明显减少,尼尔雌醇组和高剂量刺芒柄花素组较模型组明显升高(P<0.05),低剂量刺芒柄花素组较模型组也升高,但两者差异无统计学意义。real-time结果显示尼尔雌醇组ERβ含量最高,高剂量刺芒柄花素组次之,低剂量略低,模型组大鼠ERβ含量最低。结论:雌激素水平降低可使大鼠主动脉血管雌激素受体β表达减少。Objective： To study the effect of formononetin on the expression of estrogen receptor β （ERβ） in aortic arch of ovariectomized and hyperlipidemia rats. Method： In the experiment, SD rats were selected to establish the model of ovariectomized and hyperlipidemia rats followed by formononetin or estrogen treatment. Western blot and real-time PCR were used to examine the expression change of receptor β. Result： The results of western blot showed that the content of ERβ in rat＇ s aortic arch of the model group was significantly decreased compared with that of the sham operation group, and both nilestrol and high-dose formononetin could enhance the content of ERβ in rat＇ s aortic arch. Though the lower-dose formononetin could increase the content of ERβ in rat＇ s aortic arch, there was no significant difference compared with that of the model group. The results of real-time PCR were similar with those of above. Conclusion： Estrogen declining can reduce the expression level of ERβ in rat＇ s aortic arch, and appropriate dose of formononetin can increase the expression.
