摘  要:双向电泳是目前唯一能将数千种蛋白同时分离与展示的技术。以具有野生白花甜菜优质基因资源的甜菜M14品系为材料,从植株取材、样品制备、蛋白质定量、上样量、双向电泳、染色、脱色等方面优化,构建了高效、稳定的甜菜M14品系花器官蛋白质双向电泳技术平台。结果证明所获得的凝胶图谱背景清晰,对比度高,重复性好,蛋白质点边缘平滑,并且低丰度的蛋白也能够很好的显现。The two-dimensional electrophoresis is the only technology that can simultaneously separate and display thousands of proteins. The floral organ of sugar beet monosomic addition line M14 was used as experiment material, which had high-quality genetic resources derived from Beta corolliflora Zoss. A highly efficient and stable two-dimensional electrophoresis technology platform of floral organ in M14 had been built by optimization in sample prepa- ration, protein quantification, the sample volume, two-dimensional electrophoresis, dyeing and bleaching, etc. The comparative experimental results showed that the 2-D gel map had the features of clear background, high contrast, good reproducibility, smooth edges of proteins. Meanwhile, low abundance proteins also could be a good show.
