摘  要:目的通过观测电针对铝中毒运动神经元病模型兔脊髓组织NGF表达的影响作用,探讨针刺对运动神经元病的疗效与作用机理,为临床针刺治疗运动神经元病提供实验依据。方法将50只健康日本大耳白兔随机分为5组,空白对照组、模型对照组、电针治疗组、中药治疗组和针药配合治疗组,每组10只。除空白对照组外,均采用铝中毒造模方法建立运动神经元病理模型。然后,针刺治疗组行穴位电针治疗;中药治疗组服用益髓汤治疗;针药配合治疗组采用电针配合益髓汤治疗。治疗35 d后处死大白兔,打开椎管,切取脊髓组织标本分别固定后,采用原位杂交法测定脊髓组织的NGF-mRNA表达水平。结果实验结果显示模型组脊髓组织中NGF-mRNA表达降低;而电针治疗组脊髓组织中NGF-mRNA明显高于模型组,两者比较有显著性差异。结论电针对脊髓组织NGF表达有促进作用,说明电针对运动神经元病有治疗作用。Objective To probe the therapeutical effect and mechanism of electroacupuneture on treatment of motor neuron disease through observation on the effects of electroacupuncture on the expression of NGF - mRNA in RabbitsSpihal Cord, thus providing experimental basis for clinical use of electroacupuncture. Methods 50 healthy Japanese rabbits have been randomly divided into five groups, including Control Group, Model Group, Electroacupuncture Group, Herbal Group and Eleetroacupuncture - Herbal Group, with 10 rabbits in each group. Except Control Group, the other four groups have been made into pathological model of motor neuron disease by way of aluminum poisoned modeling. Electroacupuncture has been applied on Electroacupuncture Group, Yisui Decoction on Herbal Group and Electroacupuncture in Combination with Yisui Decoction on Electroacupuncture - Herbal Group. After 35d treatment, rabbits have been sacrified and their spinal cord has been opened. Following fixation of specimen from the spinal cord, in situ hybridization has been applied to assess the expression of NGF - mRNA in spinal cord. ResultsLab data reveals decrease of NGF - mRNA expression in Model Group, while the Electroacupuncture Group sees higher expression in comparison with that of the Model Group. ConclusionElectroacupuncture facilitates the expression of NGF - mRNA in spinal cord and has therapeutic effect on motor neuron disease.
