摘  要:目的:建立用替代对照品法测定重楼药材中重楼皂苷Ⅰ、重楼皂苷Ⅱ、重楼皂苷Ⅵ、重楼皂苷Ⅶ4种重楼皂苷的含量测定方法。方法:采用高效液相色谱法,以柴胡皂苷d为替代对照品,使用Waters SymmetryShield RP18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-水为流动相梯度洗脱,流速1.0 mL.min-1,检测波长203 nm,柱温27℃,同时测定重楼药材中4种重楼皂苷的含量。结果:以柴胡皂苷d与4种重楼皂苷的峰面积比值和柴胡皂苷d与4种重楼皂苷的浓度比值得到的标准曲线具有良好的线性关系,加样回收率在101.6%~104.2%之间。结论:柴胡皂苷d作为替代对照品可同时测定重楼药材中重楼皂苷Ⅰ、重楼皂苷Ⅱ、重楼皂苷Ⅵ、重楼皂苷Ⅶ4种重楼皂苷的含量。Objective:To establish a quantitative analysis method of four saponins(Chonglou saponin Ⅰ,Chonglou saponin Ⅱ,Chonglou saponin Ⅵ,Chonglou saponin Ⅶ)in Rhizoma Paridis with a substitute standard.Methods:An HPLC method was builded to determine the four saponins while saikosaponin d was used as substitute standard.The method was established by an Waters SymmetryShield RP18(4.6 mm×250 mm,5 μm)column with 27 ℃ column temperature,the mobile phase of acetonitrile-water with the gradient elution at a flow rate of 1.0 mL·min-1,and the detection wavelength of 203 nm.Results:The four calibration curves which were made by the ratio of peak areas and concentrations were in good linearities,and the recovery was between 101.6% and 104.2%.Conclusion:Saikosaponin d can be used as the substitute standard to determinate the four saponins in Rhizoma Paridis simultaneously.
