摘 要:为研究托佩克猪SLA-2基因功能,设计引物,克隆了该品系猪SLA-2基因(SLA-2-TPK)cDNA全长序列,并通过分子生物学软件分析其基因特征。结果显示,SLA-2-TPK cDNA全长为1 119 bp,其中3-1 097为编码区,共编码364个氨基酸,分别在第125、188、227和283位置出现半胱氨酸残基,含有两对链内二硫键。氨基酸同源性分析显示,SLA-2-TPK与其他SLA-2、SLA-3和SLA-1序列的同源率分别为86.0%-98.9%、85.0%-89.4%和84.2%-91.7%。系统进化树显示,SLA-2-TPK与韩国品系猪DQ883211遗传距离较近;各功能区分析,SLA-2-TPK与人HLA-A2和小鼠H-2K分子结构相似,且保留了人HLA-A2基因的部分功能位点。结果表明,SLA-2-TPK是典型的SLA-2等位基因,在进化程度上接近于部分韩国品系猪。In order to study the function of SLA-2 gene for TOPIGS pig,a pair of primers were designed to amplify the whole cDNA of SLA-2 from TPPIGS pig and then the genetic characterization of the interest gene was analyzed by computer.After cloning,sequencing and analyzing by computer,SLA-2-TPK was 1 119 bp and the 3-1 097 site was the ORF domain which coded for 364 amino acids with two sets of disulfide bond in intro-chain constituted by four cysteines situated in 125,188,227 and 283 sites.The amino acid identical ratios between SLA-2-TPK and other SLA-2,SLA-3 and SLA-1 alleles were 86.0%-98.9%,85.0%-89.4% and 84.2%-91.7%,respectively.The phylogenetic tree of the SLA-2-TPK demonstrated that it was close to some breeds of swine in Korean,such as DQ883211.Besides,the SLA-2-TPK was similar to the HLA-A2 and H-2K in functional domains and it preserved some functional sites of HLA-A2.The results indicated that SLA-2-TPK was a typical SLA-2 allele that might be closely relative to some breeds of swine in Korean.
