摘  要:目的:观察绞股蓝颗粒对早期糖尿病肾病(DN)模型大鼠肾脏一氧化氮(NO)及一氧化氮合成酶(NOS)表达的影响,探讨其治疗糖尿病肾病的作用机制。方法:48只SD大鼠,利用单侧肾切除加STZ注射法改良复制糖尿病肾病大鼠模型,随机分为正常组、模型组、治疗组(绞股蓝颗粒高、中、低剂量组及缬沙坦组),各组大鼠给予相应药物灌胃,共4周。观察各组大鼠一般情况、生化指标、肾组织NO含量、NOS活性及诱导型NOS(iNOS)mRNA表达的变化。结果:治疗组一般情况、生化指标变化均优于模型组。治疗组肾组织NO含量显著减少,NOS活性明显降低,肾组织iNOSmRNA明显下调,与模型组比较,差异有统计学意义(P<0.05或P<0.01)。结论:绞股蓝颗粒可用于治疗大鼠早期糖尿病肾病,其机制可能与调节一氧化氮及一氧化氮合成酶表达有关。Objective:To investigate the effects of gynostemma pentaphylla powder on the renal expression of nitric oxide and nitric oxide synthases in diabetic nephropathy.Methods:A total of 48 Sprague Dawley rats were divided into 6 groups:normal control group,model group,and treatment groups(high,middle,low dose groups of gynostemma pentaphylla powder and valsartan group,a 4-week treatment duration).Diabetic nephropathy model was established in rats.General state of health was observed and biochemical indicators,the content of nitric oxide and the activity of nitric oxide synthases in the kidney,and the expression of mRNA of inducible nitric oxide synthase in the kidney was measured.Results:The general state of health and biomedical examination were improved in all treatment group than those in the model group.In the treatment groups,the content of nitric oxide and the activity of nitric oxide synthases,and the expression of inducible nitric oxide synthase mRNA were lower than those of the model group(P0.05 or P0.01).Conclusion:Gynostemma pentaphylla powder may regulate the disorder of nitric oxide and nitric oxide synthases system in diabetic nephropathy.
