摘  要:[目的]观察穿龙薯蓣皂苷治疗再生障碍性贫血(简称再障)小鼠的疗效及骨髓CD3+、CD4+、CD8+的表达。[方法]建立再障小鼠模型,各组分别喂饲生理盐水、穿龙薯蓣皂苷、环孢菌素A、雷公藤多苷,连续给药14 d,取骨髓单个核细胞培养24h后,应用流式细胞术检测CD3+、CD4+、CD8+的表达水平。[结果]模型组CD3+和CD4+表达明显低于正常组(P<0.05),CD8+表达高于正常组(P<0.05),各治疗组与模型组相比均有明显差异(P<0.05),穿龙薯蓣皂苷中、高剂量组与正常组相比差异无统计学意义(P>0.05),与两阳性对照组相比差异均有统计学意义(P<0.05)。CD4+/CD8+比值:模型组低于正常组(P<0.05),中剂量组与模型组相比有明显差异(P<0.05),其余各治疗组与模型组相比均无明显差异(P>0.05)。[结论]薯蓣皂苷能调节提高再障小鼠骨髓CD3+、CD4+的表达,抑制小鼠骨髓CD8+的表达,促进CD4+/CD8+比值的恢复,从而抑制骨髓T细胞的异常激活,促进骨髓造血功能的恢复。[Objective] To observe the effect of Dioscorea nipponica Makino in treating in aplastic anemia and the expression of CD3+,CD4+,CD8+ on bone marrow mononuclear cells.[Methods] The 105 healthy ICR mice were chosen.The 15 mice were served for normal control.The other 90 mice were served for inducing aplastic anemia(AA) with Sun Jiyuan methods.After model identification,successful model mice were randomly divided into 6 groups(model group;small dosage group;middle dosage group;large dosage group;Cyclosporine A group;Tripterygium Glycoside group,n=15 each).From the 1st day to the 14th day,these mice were gavaged with different methods daily.Blood routine examination was performed on the 15th day.Bone marrow mononuclear cells for cell culture were extracted,and CD3+,CD4+,CD8+ cells were detected with Flow Cytometer after 24 hours.[Results] Compared with the model group,the expression of CD3+,CD4+,CD4+/CD8+ in treatment groups were significantly increased(P0.05).CD8+ obviously reduced,and it didn't have significant difference in normal group,middle dosage group and large dosage group(P0.05),but it had significant difference in middle dosage group and large dosage group and the two control groups.[Conclusion] Dioscorea nipponica Makino can regulate the balance of T-lymphocyte subsets in mice with AA.
