摘 要:黄芪是一种多年生草本植物,在医学上具有很高的药用价值,市场需求量很大,但是黄芪种子的萌发率很低,本文优化常温放置一年的膜荚黄芪种子的萌发条件,并诱导愈伤组织.分别用浓硫酸、H_2O_2和灭菌的自来水处理黄芪种子不同时间,检测种子萌发率.再用幼嫩的子叶在MS+1.5mg/L 2,4-D+1.0mg/L NAA的培养基上诱导愈伤组织.结果发现:用98%浓硫酸浸泡3min后黄芪种子的发芽率最高,同时获得状态很好的愈伤组织.因此,98%浓硫酸预处理可以提高黄芪种子的发芽率,愈伤组织的获得为黄芪组织培养体系的建立奠定基础.Astragalus membranaceus ( Fisch. ) Bge. is a perennial herb with a high medical value. So, demand of Astragalus is increasing in the market. But germination rate of Astragalus seed is very how. This paper talks a bout to optimize conditions of germination of Astragalus seeds which were laid in normal temperature one year. Then, callus of Astragalus was induced. Astragalus seeds were treated with sulfuric acid, H202 and water of sterilization at different time, and germination rate of Astragalus seeds was detected. Then, callus was induced with young cotyle- don in MS + 1.5 mg/L 2,4 - D + 1.0 mg/L NAA. The result showed germination rate of Astragalus seed with 98% sulfuric acid in three minutes was highest in all treatment tests . And the fine callus was obtained. Therefore 98% sulfuric acid could improve the germination rate of Astragalus seeds . The gain of Astragalus callus has established the foundation of tissue culture.
