摘 要:目的:建立云南松中莽草酸的高效液相色谱测定方法,以分别考察同一产地不同采收期的云南松中莽草酸的含量。方法:色谱柱为Agilent Eclipse XDB-C18(Φ4.6 mm×150 mm,5μm),流动相为甲醇-1%磷酸水溶液(10∶90),流速0.8 mL/min,检测波长214 nm,柱温25℃。结果:莽草酸在0.0312~0.4368 ug范围内与峰面积呈良好的线性关系(r=0.9999),平均加样回收率为99.97%,RSD为1.06%(n=6)。结论:此方法准确、简便、重复性好,可用于含莽草酸的药材及制剂的质量控制。Objective: To establish HPLC method for the determination of shikimic acid in Pinus yunnanensis Franch and inspect the content of the shikimic acid in Pinus yunnanensis Franch harvested in different periods in the same region. Method: The column was Agilent Eclipse XDB-C18 (Φ4.6 mm×150 mm, 5 μm); the mobile phase was methanol-1% phosphoric acid (10∶90), with 0.8 mL/min of flow rate, 214 nm of detection wavelength, and 25 ℃ of column temperature. Results: The shikimic acid showed a good linear relationship (r=0.9999) at the range of 0.0312~0.4368 ug peak area. The average recovery rate was 99.97%, RSD of 1.06% (n=6). Conclusion: This method is accurate and simple with good repeatability and can be used to control the quality of shikimic acid-containing herbs and preparations.
