摘　　要：利用线粒体COⅡ部分基因序列对直突摇蚊亚科代表性属级阶元进行分子系统学研究。共测定24属30种基因序列,利用ClustalX软件对比并用MEGA4.1软件分析其碱基组成,以Chironomus samoensis Edwards作为外群构建分子系统树。结果表明:拟开氏摇蚊属Parakiefferiella和克莱施密摇蚊属Krenosmittia,伪直突摇蚊属Pseud-orthocladius,陆直突摇蚊属Georthocladius各自互为姐妹群,此结果与基于形态学的系统发育研究结果相一致。毛突摇蚊属Chaetocladius和直突摇蚊属Orthocladius、拟环足摇蚊属Paracricotopus和施密摇蚊属Smittia、真开氏摇蚊属Eukiefferiell和肛脊摇蚊属Mesosmittia在本实验结果中得到较高的支持率形成姐妹群关系,但因系统发育关系尚无前人研究而有待做进一步验证。本研究同时证明线粒体COⅡ基因片段在分析摇蚊科属间及属内种间关系上具有一定的指导意义。In this paper,the molecular phylogenetic analysis on the generic level of Orthocladiinae has been studied based on partial sequences of mitochondrial COⅡ gene.This gene fragment of 30 species from 24 genera of the Orthocladiinae and outgroups were sequenced.Sequences were aligned using Clustal X 1.83.Sequence statistics were obtained by MEGA 4.1.Phylogenetic analysis under the Neighbor-joining（NJ） and Minimum Evolution（ME） were performed as implemented in MEGA 4.1.The result showed that：The genera of Parakiefferiella and Krenosmittia,Pseudorthocladius and Georthocladius were sister groups respectively.These phylogenetic relationships have been supported by morphologic character.The genera of Chaetocladius and Orthocladius,Paracricotopus and Smittia,Eukiefferiell and Mesosmittia were formed sister groups respectively,with well bootstrap value.There was no phylogenetic study of the subfamily Orthocladiinae,so the phylogenetic relationship of these genera within the Orthocladiinae is needed to be confirmed in the future.According to the result of this paper,the CO Ⅱ gene can provide considerable phylogenetic information on the level of inter and intra genera in Chironomidae.
