摘 要:目的观察包裹脑源性神经营养因子(brain derived neurotrophic factor,BDNF)的壳聚糖/海藻酸钠微囊对大鼠视神经损伤的保护作用。方法在视神经损伤模型的大鼠玻璃体腔内注入包裹BDNF的壳聚糖/海藻酸钠微囊。第一组中对视神经作单次损伤,第二组中两次损伤视神经。通过视网膜神经节细胞(retinal ganglion cells,RGCs)密度计数,评价包裹BDNF的壳聚糖/海藻酸钠微囊对RGCs的保护作用。结果 BDNF微囊注入组和游离BD-NF注射组,在注入后14天和28天组与空白PBS及壳聚糖/海藻酸钠微囊组比较RGC密度差异均有统计学意义(P<0.01),BDNF微囊注入组和游离BDNF注射组的RGCs的密度高于空白对照组。单次损伤视神经的BDNF微囊注入组与游离BDNF注射组的两组间RGC密度差异无统计学意义(P>0.05)。而两次损伤视神经的BDNF微囊注入组与游离BDNF单次注射组的两组间RGC密度差异有统计学意义(P<0.01),BDNF微囊注入组的RGCs的密度高于游离BDNF单次注射组。BDNF微囊注入组与BDNF两次注射组的两组间RGC差异无统计学意义(P>0.05)。结论对于受到反复损伤的RGCs,BDNF微囊释放的BDNF在注入后对损伤下的RGCs起到了明显的保护作用,优于单次注入BDNF。而对只受到单次损伤的RGCs,BDNF微囊释放的BDNF与单次注入BDNF效果相同。Objective To evaluate the effect of chitosan/alginate microcyst encapsulating BDNF via intravitreal injection in SD rats with damaged optic nerve(ON).Methods In one group,SD rats in which ON was damaged once,underwent intravitreal injection of chitosan/alginate microcyst encapsulating BDNF or free BDNF or vehicle.In another group,SD rats,in which ON was damaged twice,underwent intravitreal injection of chitosan/alginate microcyst encapsulating BDNF or BDNF or vehicle too.To determinte RGCs densities,cells were labled retrogradely with granular blue.Results The averange RGCs densities of microcyst encapsulating BDNF group and free BDNF group were higher than those of vehicle group(P0.01).In ON once damaged group,the averange RGCs densities of microcyst encapsulating BDNF group had no statistic diference compared with those of free BDNF group(P0.05).In ON twice damaged group,the averange RGCs densities of microcyst encapsulated BDNF group were higher than those of free BDNF single injection group(P0.01).However,no statistic diference were formed compared with free BDNF twice injection group(P0.05).Conclusion BDNF released by microcyst can protect RGCs under repeated damage and its protecting effect is stronger than single injection of free BDNF.BDNF release by microcyst has the same protecting effect to RGCs undergone single damage compared with free BDNF injection.
