杀镰刀菌素对枯草杆菌致死作用的研究

摘  要:通过检测OD600、抑菌圈和发酵液澄清度的变化,考察了杀镰刀菌素对枯草杆菌的抑制杀菌作用。实验利用HPF荧光证实杀镰刀菌素致使枯草杆菌细胞内产生氢氧自由基(.OH),添加抗坏血酸(Vc)减少.OH的形成,提高枯草杆菌的存活率。应用基因芯片对5min转录变化分析发现sigW调控的基因及胞膜相关基因高表达,用透射电镜观察发现出现空囊体及大量细胞碎片。Through observing the changes of OD600, inhibition zone and the clarity of culture, the inhibition activity of fusaricidin to Bacillus subtilis was investigated. The HPF experiment verified that the formation of . OH could be induced by fusaricidins in Bacillus subtilis. The results showed that the production of . OH would he reduced and the survival rate of Bacillus subtilis increased when aseorbic acid was added in the broth with fusaricidins, Geneehip was used to study the transcript profiles of Bacillus subtilis in response to fusaricidins at 5 min. After adding fusaricidins,Bacillus subtilis produced lots of cell components associated with membrane, and the genes regulated by sigW were induced. Through observing by transmission electron microscop ( TEM), a lot of thylakoid and cell debris were discovered.
