摘 要:目的探讨八氯代二苯并二噁英(1,2,3,4,6,7,8,9-octachlorodibenzo-p-dioxin,OCDD)对小鼠心脏基因表达谱改变的影响。方法将20只7~8周龄SPF级雄性C57BL/6j小鼠按体重随机分为4组,分别为对照(玉米油)组和低(1.25×10-6 g/ml)、中(1.25×10-5 g/ml)、高(1.25×10-4 g/ml)浓度OCDD染毒组,每组5只。采取灌胃方式进行染毒,染毒容量为8 ml/kg,每天1次,连续染毒30 d。应用基因芯片技术比较不同浓度OCDD染毒组和对照组雄性小鼠心脏基因表达差异。结果共检测了小鼠心脏基因41 253个。与对照组相比,低浓度OCDD染毒组和对照组差异基因总数5 490个,其中,上调2 257个,下调3 233个;中浓度OCDD染毒组和对照组差异基因数2 776个,其中,上调941个,下调1 835个;高浓度OCDD染毒组和对照组的差异基因数14 911个,上调10 476个,下调4 435个。各浓度OCDD染毒组和对照组共同的2倍以上差异基因为288个。结论亚慢性OCDD染毒对小鼠的心脏相关基因表达造成影响,提示心脏可能是OCDD毒性作用的靶器官。Objective To investigate the effects of 1,2,3,4,6,7,8,9-octachlorodibenzo-p-dioxin (OCDD) on the cardiac gene expression profile in the heart of mouse. Methods Twenty male C57BL/6j mice aged 7 or 8-weeks, SPF class, were randomly divided into four groups, 5 mice in each, as normal control group(maize oil),low concentration group( 1.25×10^-6 g/ml, OCDD), moderate concentration group (1.25 × 10^-5 g/ml, OCDD)and high concentration group ( 1.25 ×l0^-4 g/ml, OCDD). Pretreatment groups were given solvents by 8 ml/kg each day in the way of garage for 30 days. The differences of gene expression profile between OCDD exposed groups and control group in C57BL/6j male mice were compared by gene chip technology. Results A total of 41 253 heart genes were detected. Compared with the control group,in low OCDD exposure group, the number of different genes was 5 490,with 2 257 up-regulated and 3 233 down-regulated; in moderate OCDD exposure group, it was 2 776 ,with 941 up-regulated and 1 835 down-regulated; In high OCDD exposure group, it was 14 911, with 10 476 up-regulated and 4 435 down-regulated. The number of the different genes of more than two times both in OCDD exposure and the control groulSs was 288. Conclusion The effect of sub-chronic exposure to OCDD on the cardiac gene expression in male C57BL/6j mice indicates that the heart is probably the target organ of OCDD.
