摘 要:目的探讨三七总苷对二甲基亚硝胺(DMN)致肝纤维化模型大鼠的干预作用。方法 60只SD大鼠,随机选取10只作为正常组,其余50只以DMN复制中度肝纤维化模型,成功造模37只,选取8只作为模型组,剩余随机分为三七总苷组(8只)、干扰素组(8只)、三七总苷+干扰素组(7只)、生理盐水组(6只)。三七总苷组给予血栓通冻干粉1.5mg/100g、干扰素组给予注射重组人干扰素γ8.675mg/100g、三七总苷+干扰素组给予以上两种药物、生理盐水组给予等量生理盐水,以上均每日肌肉注射1次,连续给药8周后检测各组大鼠血清肝功能、透明质酸,结合常规病理HE、苦味酸-天狼星红染色,评价肝组织炎症及纤维化程度。结果正常组及各干预组与模型组血清总蛋白(TP)、白蛋白(Alb)、丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)和血清透明质酸比较差异均有统计学意义(P<0.05);各组分别与模型组大鼠肝纤维化分级半定量后分值比较差异均有统计学意义(P<0.05);三七总苷组、干扰素组、三七总苷+干扰素组SSS半定量计分分别与模型组比较,差异均有统计学意义(P<0.05)。结论三七总苷能够改善DMN所致肝纤维化大鼠肝功能,降低血清透明质酸含量,改善肝纤维化病理分级。Objective To observe the intervention effect of notoginseng total glucoside on the dimethylnitrosamine (DMN) induced hepatic fibrosis of rat models. Methods Of the 60 SD rats, 10 were randomly selected and put into the normal control group, the rest was made models of hepatic fibrosis to a medium degree induced by DMN. From the 37 successfully made models, 8 randomly selected were taken as the model control group. The rest of rats was randomized into notoginseng total gluco-side group, 8 rats, interferon group, 8 rats, notoginseng total glucoside -k interferon group, 7 rats, and physiological saline negative control group, 6 rats. The muscular injection with Freeze-dried Powder Xueshuantong for Injection (Xueshuantong) 1.5mg/100g, Recombinant Human Interferon γ for Injection (INF-γ) 8. 675mg/lOOg, both Xueshuantong and INF-γ, and the physiological saline in the same dosage was given respectively, once a day, 8 weeks in succession. The effect on the inflamma- tion and fibrosis degrees of hepatic tissues was evaluated by serum assaying on the indices of liver function and hyaluronic acid (HA) and the results of routine pathological HE and Sirius Red Stains examinations. Results The comparison of TP, AIb, ALT, AST, and HA in the normal control group and all intervention groups with the model control group showed a statistical difference (P〈0. 05), the comparison in the model control group with the negative control group showed a statistical difference (P〈0.05), the comparison of the fibrosis score formed by the Semi Scoring System (SSS) of all groups with the model control group respectively showed a statistical difference (P〈0, 05), and the comparison of the SSS scores of the notoginseng total glucoside group, interferon group, and the notoginseng total glucoside + interferon group with the model control group respectively showed a statistical difference (P〈0.05).
