摘 要:目的探讨大黄虫丸对大鼠肺纤维化形成阶段神经递质的干预作用。方法普通级Wistar大鼠36只,随机分为正常组、模型组、大黄虫丸组各12只,除正常组外,其余2组均采用气管内注入平阳霉素复制肺纤维化模型,大黄虫丸组灌胃浓度为80mg/ml药液1.5ml/100g体重,模型组灌胃等量蒸馏水,每天给药1次,各组于造模后第28天处死,比较各组大鼠肺、下丘脑及海马组织中多巴胺(DA)、去甲肾上腺素(NE)、5-羟色胺(5-HT)含量。结果模型组大鼠肺与下丘脑组织中NE、DA、5-HT及海马组织中5-HT含量均与正常组有统计学差异(P<0.05);大黄虫丸组肺、下丘脑NE、DA、5-HT含量及海马组织中NE、5-HT含量均与模型组有统计学差异(P<0.05)。结论肺纤维化形成阶段肺与下丘脑DA、NE、5-HT及海马组织中5-HT含量升高,大黄虫丸能调节肺纤维化模型大鼠肺及脑组织中NE、DA、5-HT的含量。Objective To explore the intervention effect of Dahuang Zhechong Pills [Pills containing Dahuang (Rhubarb) and Tubiechong (Ground Beetle) for dissolving blood stasis] on neurotransmitter in the formation stage of pulmonary fibrosis of rats. Methods Female Wistar rats were randomized into normal group, model group, and Dahuang Zhechong Pills group, with 12 in each. Except the normal group, the other rats were made pulmonary fibrosis models through endotracheal injection with Pingyangmycin (PYM). The Dahuang Zhechong Pills group was treated with Dahuang Zhechong Pills solution in the density of 80mg/ml according to 1.5ml/100g body weight for intragastric feeding, while the model group was given the same amount distilled water for intragastric feeding, once a day. On the 28th day after modeling, the rats were put to death. The contents of dopamine (DA), norepinephrine (NE), and serotonin (5-HT) in the lung, hypothalamus, and hippocampus tissues of rats of all groups were compared with each other. Results The contents of NE, DA, and 5-HT in the lung and hypothalamus tissues and 5-HT in the hippocampus tissues of the rats of model group were statistically different from those of the normal group (P〈0.05). The contents of NE, DA, and 5-HT in the lung and hypothalamus tissues and NE, 5-HT in the hippocampus tissues of the rats of Dahuang Zhechong Pills group were statistically different from those of the model group (P〈0.05). Conclusion In the formation stage of pulmonary fibrosis, the contents of DA, NE, and 5-HT in the lung and hypothalamus tissues and 5-HT in the hippocampus tissues are increased. Dahuang Zhechong Pills is effective in adjusting the contents of DA, NE, and 5-HT in the lung and brain tissues of rat models of pulmonary fibrosis.
