摘 要:目的探讨黄连解毒汤(Huanglian Jiedu Decoction,HJD)及其拆方含药血清对前脂肪细胞增殖和分化的影响及其机制。方法制备HJD及其拆方的大鼠含药血清;培养3T3-L1前脂肪细胞;以四甲基偶氮唑盐(methyl thiazolyl tetrazolium,MTT)法检测3T3-L1前脂肪细胞的增殖;油红O染色并通过比色定量分析检测3T3-L1前脂肪细胞分化过程中胞浆脂质的堆积;采用逆转录聚合酶链反应技术检测过氧化物体增殖物活化受体γ(peroxisome proliferator activated receptorγ,PPARγ)和CAAT/增强子结合蛋白α(CAAT/enhancerbinding protein,C/EBPα)mRNA的表达。结果与空白对照组比较,HJD、黄芩栀子合煎和栀子的含药血清对3T3-L1前脂肪细胞增殖均表现出明显的促进效应(P<0·05,P<0·01);HJD、黄芩栀子合煎、黄连和栀子的含药血清处理前脂肪细胞,分化后胞浆中脂滴明显减少,PPARγ和C/EBPαmRNA表达明显降低,与空白对照组比较,差异有统计学意义(P<0·05,P<0·01)。结论 HJD促进前脂肪细胞的增殖,减少脂肪细胞分化过程中脂质的堆积,抑制脂肪细胞的分化,可能与其降低PPARγ和C/EBPαmRNA表达有关;HJD发挥此作用的主要成分可能来自于黄芩栀子药对。Objective To study the effects and mechanisms of Huanglian Jiedu Decoction(HJD) and its disassembled recipes containing serums on the proliferation and differentiation of preadipocytes.Methods HJD and its disassembled recipes containing serums were prepared.The 3T3-L1 preadipocytes were cultured.The proliferation of 3T3-L1 preadipocytes was detected by methyl thiazolyl tetrazolium(MTT) method.The accumulation of lipid droplets in the cytoplasm of differentiated preadipocytes was observed by oil red O staining and quantitatively analyzed by colorimetry.The mRNA expressions of peroxisome proliferation activated receptor γ(PPARγ) and CAAT/enhancer binding protein(C/EBPα) were detected by reverse transcription polymerase chain reaction(RT-PCR).Results Intervention with serum containing HJD,Phellodendron amurense Rupr plus Gardenia jasminoides Ellis,or Gardenia jasminoides Ellis showed significantly stimulative effects on the proliferation of preadipocytes,as compared with that in the blank control group(P0.05,P0.01).The preadipocytes treated with serum containing HJD,Phellodendron amurense Rupr plus Gardenia jasminoides Ellis,Coptis chinensis Franch or Gardenia jasminoides Ellis showed that the lipid droplets in the cytoplasm were significantly lessened,so did the mRNA expressions of PPARγ and C/EBPα when compared with the blank control group(P0.05,P0.01).Conclusions HJD promoted the proliferation of preadipocytes,decreased the accumulation of lipid droplets during the differentiation of adipocytes,and inhibited the differentiation of adipocytes,which might be associated with its effects on decreasing the mRNA expressions of PPARγ and C/EBPα.Phellodendron amurense Rupr and Gardenia jasminoides Ellis were the main components of HJD playing these roles.
