摘  要:目的建立关黄柏、黄柏及其AB-8型大孔树脂精制提取物的HPLC指纹图谱,研究饮片与提取物指纹图谱的相关性。方法采用zorbax SB C18色谱柱;流动相为乙腈-0.4mol·L-1的氯化铵溶液(梯度洗脱);检测波长为230nm;柱温为40℃;流速为1.0mL·min-1。结果关黄柏标示出17个共有峰,关黄柏提取物能检出关黄柏中的15~17个共有峰,鉴别了4个特征峰;10批关黄柏及其提取物的相似度分别为0.992~0.999、0.964~0.999,同批次关黄柏与提取物之间的相似度为0.926~0.999。黄柏及其提取物均标示出与关黄柏相同的14个共有峰,但相对峰面积比值相差较大;3批黄柏及其提取物的相似度分别为0.999~1.000、0.994~1.000,同批次黄柏与提取物之间的相似度为0.988—0.989。关黄柏与黄柏、关黄柏提取物与黄柏提取物指纹图谱模式之间的相似度分别为0.807、0.815。结论该方法准确可靠,重复性好,为关黄柏、黄柏及其提取物的质量控制提供了方法依据,同时为含关黄柏复方制剂的工艺与质量控制提供了参考。Objective To establish the method of fingerprint analysis of Cortex Phellodendri Amurensis and Cortex Phellodendri Chinese by HPLC, and to study the correlation of fingerprints of their decoction pieces with those of refined extract. Methods HPLC was used with the Zorbax SB C18 column, the acetonitrile-ammonium chloride solution (c =0.4 mol·L-1, in gradient elution) was employed as a mobile phase, detection wavelength was 230 nm, column temperature was 40 ℃, and flow rate was 1.0 mL min-l. Results Seventeen common peaks were separated in the decoction pieces of Cortex Phellodendri Amurensis and 15 -17 common peaks in the refined extract of Cortex Phellodendri Amurensis, and 4 peaks were identified. The similarities of 10 batches of the decoction pieces were 0.992- 0.999 and the similarities of refined extract were 0.964- 0.999. The similarities between the decoction pieces and refined extract were 0.926-0.999. In Cortex Phellodendri Chinese, 14 common peaks were separated, the similarities of 3 batches of the decoction pieces and refined extract were 0.999- 1.000 and 0.994- 1.000, respectively, and the similarities between the decoction pieces and refined extract were 0.988- 0.989. Conclusion The method is accurate and reproducible, which can be used for the quality control of decoction pieces and refined extract of Cortex Phellodendri Amurensis and Cortex Phellodendri Chinese as well as for compound preparation containing Cortex Phellodendri Amurensis.
