Differential Expression of Transient Receptor Potential Channel A1 in Different Parts of the Sensory Pathway in Inflamed Rat Urinary Bladder

摘  要:目的瞬时受体电位蛋白通道A1(TRPA1)参与机械性感觉、痛觉的传导以及炎性反应,我们制作大鼠的膀胱炎症模型,检测在膀胱传入通路中不同部位TRPA1的表达情况。方法采用大鼠腹腔内注射环磷酰胺制作膀胱炎症模型,正常大鼠作为对照组。采集膀胱黏膜层及脊髓后根神经节,利用实时定量PCR检测TRPA1转录水平的变化。结果大鼠腹腔内注射环磷酰胺2 d后,肉眼及病理检查证实膀胱炎性反应。TRPA1在膀胱黏膜层的表达水平没有明显变化,在脊髓后根神经节中,与对照组相比,炎症组的TRPA1的表达水平明显上调(P<0.05)。结论在膀胱炎症状态下,TRPA1在黏膜层的表达没有明显变化,提示它作为黏膜层的机械敏感性受体参与炎症诱发的排尿改变的可能性较小;TRPA1在脊髓后根神经节中表达上调,感觉神经元表达的TRPA1可能参与膀胱炎性反应,并且与膀胱炎时病理性的排尿方式相关。Objective Transient receptor potential channel A1(TRPA1) is a candidate for mechanosensory transduction,nociception,and also involved in inflammation.We made urinary bladder inflammation model in rats,then investigated the expression level of TRPA1 in different parts of bladder sensory pathway.Methods Bladder inflammation model was made by injection of cyclophosphamide intraperitoneally in rats.The bladder mucosa and dorsal root ganglia(DRG) innervating the urinary bladders were harvested.The transcription level of TRPA 1 was examined by real-time polymerase chain reaction.Results The inflammation of bladder 2 days after injection of cyclophosphamide was confirmed microscopically.In bladder mucosa,there was no obvious difference in TRPA1 mRNA level between inflammation and normal groups.In DRG,the TRPA1 mRNA level was significantly up-regulated compared with control group(P 0.05).Conclusion TRPA1 expression level showed no difference in mucosa in inflammation group,indicated that TRPA1 in mucosa was less likely to be involved in the abnormal micturition or inflammation,especially as a mechanosensory receptor.However,after inflammation was induced,TRPA1 was up-regulated in the DRG innervating the urinary bladder,which indicated that TRPA1-expressing primary sensory nerves was probably related with either the inflammation or the changes in micturition pattern.
