糖尿病大鼠膀胱平滑肌组织中AGEs与iNOS表达的相关性研究

摘  要:目的探讨糖尿病大鼠膀胱平滑肌组织糖基化终产物(AGEs)、诱导型一氧化氮合酶(iNOS)的表达变化及他们表达的相关性。方法 64只雄性Wista大鼠随机分为糖尿病模型组(DM组)和对照组(NC组),每组32只。在DM组大鼠模型制备成功后的4、8、12和20周,2组各杀检8只大鼠。使用免疫组化法观察膀胱平滑肌组织中AGEs与iNOS的表达定位同时分析其含量变化。结果大量AGEs阳性细胞分别出现于4、8、12和20周DM组大鼠膀胱平滑肌组织中,其累计光密度值(IOD)高于NC组,并随病程进展而增高(P<0.01);4周末DM组大鼠膀胱平滑肌组织中可见少量iNOS阳性细胞,NC组的大鼠也可见少量iNOS阳性细胞,两组比较IOD值差异无统计学意义(P>0.05);8、12和20周DM组大鼠膀胱平滑肌组织中可见大量iNOS阳性细胞,IOD值高于相应阶段的NC组(P<0.01),并随病程进展而逐渐增高(P<0.01);Pearson相关分析显示:DM组大鼠膀胱平滑肌组织中AGEs与iNOS的表达水平呈显著正相关(r=0.897,P<0.01)。结论糖尿病大鼠膀胱平滑肌组织中AGEs和iNOS的表达正相关,AGEs对iNOS的表达可能存在诱导作用。Objective To study the changes of the expressions of advanced glycation end products(AGEs) and inducible nitric oxide synthase(iNOS) in the diabetic rats smooth muscles of bladder and the relationship of the expressions of AGEs and iNOS.Methods 64 male Wista rats were distributed randomly into groups of DM and NC(n = 32 each).DM models were made by STZ injection.Eight rats in both groups were killed and examined at week 4,8,12 and 20 respectively after the DM model rats were made successfully.Changes of the expressions of AGEs and iNOS in the smooth muscles of bladder were observed and analyzed by immunohistochemical method.Results In DM groups,lots of AGEs positive cells could be seen,and increased following the diabetic course.The integrated option density(IOD) of the positive cells were significantly higher than that of the corresponding stage of NC groups(P 0.01),and decreases significantly with the progression of the diabetic course(P 0.01) were seen;in the diabetic 4 weeks-subgroup,a small amount of iNOS positive cells could be seen,and in normal control 4 weeks-subgroup,a small amount of positive cells also could be seen.The diabetic 4 weeks-group compared with the normal control 4 weeks-subgroup,the IOD of the positive cells were not statistically different(P 0.05).In diabetic 8,12 and 20 weeks-subgroups,lots of positive cells could be seen,the IOD of the positive cells were significantly higher than that of the corresponding stage of NC groups(P 0.01),and the number increased gradually following the diabetic course(P 0.01).Pearson′s correlation analysis showed a positive relationship of AGEs with iNOS(r = 0.897,P 0.01) in diabetic group.Conclusion The expressions of AGEs and iNOS were markedly increased and closely correlated with each other in the smooth muscles of bladder,AGEs might be the factor which induced iNOS in the smooth muscles of bladder.
