摘 要:目的探讨酸洗脱血小板表面HLA-Ⅰ类抗原对血小板活化和凋亡的影响。方法将10(人)份健康人机采血小板(0.5 ml/份)用pH3柠檬酸缓冲液处理(同时以PBS处理作对照组)后,利用多色流式细胞技术检测血小板表面HLA-Ⅰ类抗原及P-选择素(CD62p)的表达,Annexin V染色检测血小板的早期凋亡率。结果 pH3柠檬酸缓冲液及PBS处理后的血小板表面HLA-Ⅰ类抗原的表达为(23.83±9.93)%vs.(98.15±2.20)%(P<0.05),CD62p的表达为(45.94±10.13)%vs(20.29±4.24)%(P<0.05),血小板的早期凋亡率为(65.29±15.74)%vs(14.65±5.09)%(P<0.05)。结论 pH3柠檬酸缓冲液处理血小板可有效去除血小板表面HLA-Ⅰ类抗原,并同时诱导血小板的活化和凋亡。Objective To investigate the effect of acid eluting HLA-class I antigens from the surface of platelets on platelet activation and apoptosis. Methods Ten apheresis platelets samples (0. 5 ml/sample) from healthy volunteers were treated by the citric acid solution at pH 3.0( PBS treatment as control group). The expression of HLA-class I antigens and P-selectin(CD62P) on platelet surface were analyzed by multicolor flow cytometry. The proportion of early apoptotic platelets was detected by Annexin V staining. Results After treatment with the citric acid solution and PBS,the expression of HLA- class I antigens on platelet surface was (23.83 ±9. 93)% vs (99. 34 ± 1.05)% (P 〈0. 05) ,the expression of CD62P on platelet surface was (45.94 ± 10. 13) % vs (20.29 ± 4. 24 ) % ( P 〈 0.05 ), the proportion of early apoptotic platelets was ( 65.29 ± 15.74 ) % vs ( 14. 65 ± 5.09 ) % ( P 〈 0. 05 ). Conclusion Treatment of platelets with the citric acid solution at pH 3.0 can effectively eliminated HLA class - I antigens from platelet surface, but significantly induced platelets activation and apoptosis.
