摘 要:目的:观察和探讨宫内感染致脑损伤仔鼠脑神经髓鞘改变及丰富康复训练的影响和机制。方法:连续2天对孕第17天的脂多糖(LPS)组孕鼠腹腔注射LPS(450μg/kg.d),制备脑损伤模型;对照组孕鼠腹腔注射等量无菌生理盐水。LPS组仔鼠随机分为非干预组和丰富康复训练组,丰富康复训练组于第2天开始进行干预,用电子显微镜观察d14、d28各组仔鼠脑神经髓鞘厚度情况,并应用体视学技术测量和计算脑神经髓鞘厚度平均值,同时用免疫组化方法检测仔鼠脑皮质区BDNF表达情况。结果:非干预组、丰富康复训练组仔鼠脑神经髓鞘厚度平均值均明显低于对照组,差异有显著性(P<0.01),丰富康复训练组仔鼠脑神经髓鞘厚度平均值高于非干预组,差异有统计学意义(P<0.01);丰富康复训练组仔鼠脑的皮质区BDNF阳性细胞计数明显高于对照组和非干预组,差异有显著性(P<0.01)。结论:(1)宫内感染可致脑损伤仔鼠脑神经髓鞘厚度平均值降低。(2)丰富康复训练可提高脑损伤仔鼠脑神经髓鞘厚度平均值。(3)丰富康复训练可增强脑损伤仔鼠脑组织BDNF阳性表达。Objective: To observe and investigate the changes of cerebral nerve myelin sheath in the brain of the young offspring of rats with intrauterine infection and the effects of enriched rehabilitation training. Methods: To prepare the brain damage models, injecting LPS (450 μg/kg·d)to abdominal cavity of pregnant rats in 17d and 18d; inject equally sterile NS to abdominal cavity of contrasting pregnant rats. The LPS young offspring of rats were randonfly divided into the non-intervention groups and the enriched rehabilitation training groups. The enriched rehabilitation training groups start to train in the second day. Using dectromnicroscopy to observe the cerebral nerve myelin sheath thickness in d14,d28 and stereological technique to measur and calculate the average thickness of cerebral nerve myelin sheath.Meanwhile, adopting immunohistochemical characterization of experimental technique to detect the positive expression of corticocerebra] 13DNF. Results: The results of stereological measurement in brain nerve myelin sheath thickness: the non-intervention groups and the enriched rehabilitation training groups have a lower average thickness measurements than the control groups in brain nerve myelin sheath, the difference was significant (P〈0.01); the enriched rehabilitation training groups has a higher average thickness measurements than the non-intervention groups, the difference was statistically significant (P 〈0.01). The enriched rehabilitation training groups was higher about positive corticocerebral BDNF cell count than the non-intervention groups (P〈 O.Ol).Conclutions:l. Intrauterine infection can depress the average thickness measurements of cerebral nerve myelin sheath about neonatal rats with brain damage;2. Enriched rehabilitation training can elevate the average thickness measurements of brain nerve myelin sheath about renascent rats with brain damage.3. Enriched rehabilitation training can increase the expression of BDNF in neonatal rat with brain damage.
