复方瑞康欣对吗啡依赖SK-N-SH细胞NOS活性和NO浓度的影响

摘  要:目的:观察复方瑞康欣对吗啡依赖的SK-N-SH细胞各型NOS活性和NO浓度的影响并探讨其机制。方法:SK-N-SH细胞周期同步化后,用吗啡(10μmol/L)作用48h,然后用复方瑞康欣含药血清干预12h,再用纳洛酮(10μmol/L)作用10min后,检测NOS活性和NO含量。结果:复方瑞康欣含药血清能抑制总NOS和cNOS活性(P<0.01),从而降低NO含量(P<0.01),但对iNOS活性没有影响(P>0.05)。结论:复方瑞康欣的戒毒作用与NOS活性和NO水平有关。Objective:To investigate the effect and mechanism of Compound Ruikangxin on morphine-dependent human neuroblastoma SK-N-SH cell line.Method:After cells being synchronized in free-serum culture medium for 24 hours,and then cultured in medium containing morphine(10μmol/L)for 48 hours,the cells turned into morphine-dependent cells.The morphine-dependent cells were administrated by serum with Compound Ruikangxin for 12 hours and then were precipitated by naloxone.The activity of nitric oxide synthase(NOS)and the concentration of nitric oxide(NO)were measured.Results:The three dosages of Compound Ruikangxin could inhibit the activity of cNOS(P0.01) and decrease concentration of NO(P0.01).Conclusion:The detoxification effect of Compound Ruikangxin might be linked with the activity of cNOS and the level of NO.
