摘 要:从玉米弯孢叶斑病菌中克隆到MAPK基因之一的Clm1,其与玉米小斑病菌中的ChMps1基因同源。采用InversePCR和RACE技术获得了该基因的全序列及其所在区域的旁侧序列。该基因cDNA全长1.251 kb,编码416个氨基酸。通过农杆菌介导的转化方法对其进行了敲除。Clm1敲除突变体在细胞壁完整性,分生孢子产量及形态方面发生明显变化,其侵染离体叶片的能力明显下降,纤维素酶活性有所下降,说明该基因与弯孢叶斑病菌细胞壁完整性及致病性有着密切的联系。A full-length of Clm1 gene from Curvularia lunata was cloned,which was highly homologous to the mitogen-activated protein(MAP) kinase ChMps1 in Cochliobolus heterostrophus.The whole sequence and flanking regions of Clm1 was amplified by Inverse PCR and RACE.Its full-length cDNA is 1.251 kb,encoding 416 amino acid residues.The targeted disruption of Clm1 was obtained by Agrobacterium-mediated transformation.Furthermore,it was found that the Clm1 mutants had impaired cell wall and caused low production of conidiospores,deformed spore formation and decreased disease symptoms on maize leaves.In addition,the Clm1 mutants also had an effect on the activity of cellulase.The results above were indicated that Clm1 played an important role in the cell wall integrity and pathogenicity in C.lunata.
