摘 要:本试旨在研究甲状腺素、胰高血糖素(PG)和表皮生长因子(EGF)对肉鸡小肠Ⅰ型肽转运载体(PepT1)mRNA表达量的影响。选择体重无差异的25日龄雄性肉仔鸡30只,分为5组,每组6只鸡,分别接受不同激素处理,每天1次,连续5d。对照组、EGF组、PG组分别皮下注射蒸馏水0.20mL/kg、EGF0.03m g/kg、PG0.10m g/kg;T4组、TH组分别灌服左旋甲状腺素钠(T4)330.00μg/kg、甲巯咪唑25.00m g/kg,同时皮下注射蒸馏水0.20mL/kg。最后1次处理12h后,测定血清人表皮生长因子(h-EGF)、三碘甲状腺原氨酸(T3)、PG含量以及十二指肠、空肠、回肠PepT1mRNA表达量。结果表明:皮下注射EGF和PG对血清中相应含量无显著影响(P>0.05);口服T4、甲巯咪唑分别显著提高、显著降低血清T3含量(P<0.05)。皮下注射PG(P<0.05)和口服T4(P<0.01)均显著提高了鸡小肠PepT1mRNA表达量,皮下注射EGF和口服甲巯咪唑有降低PepT1mRNA表达量的趋势,但均未达到显著水平(P>0.05)。结果提示,甲状腺素和PG在肉鸡小肽的吸收中发挥着重要调控作用。The study was conducted to investigate the effect of thyroxin(T4),pancreas glucagon(PG) and epidermal growth factor(EGF) on the mRNA expression level of oligopeptide transporter 1(PepT1) in small intestine of broilers.Thirty broilers aged 25 days with similar body weight were randomly divided into 5 groups with 6 broilers in each group.Broilers from different groups were treated with different hormones once a day for 5 days.Broilers in control group,EGF group and PG group were subcutaneously injected distilled water at 0.20 mL/kg,EGF at 0.03 mg/kg and PG at 0.10 mg/kg,respectively;those in T4 group and TH group were drenched levothyoxine Na(T4) at 330.00 μg/kg and methimazole at 25.00 mg/kg,respectively,and subcutaneously injected distilled water at 0.20 mL/kg.After 12 h of the last treatment,the human epidermal growth factor(h-EGF),triiodothyronine(T3) and PG contents in serum were determined,as well as the mRNA expression level of PepT1 in duodenum,jejunum and ileum.The results showed as follows: there was no significant difference between the EGF group and PG group on the corresponding contents in serum(P0.05);orally taken of T4 and methimazole significantly increased and decreased serum T3 content,respectively(P0.05).The mRNA expression level of Pept1 in small intestine was significantly increased by subcutaneous injection of PG(P0.05) and orally taken of T4(P0.01),but decreased by subcutaneous injection of EGF and orally taken of methimazole(P0.05).The results indicate that thyroxin and PG play import roles in regulation of peptides absorption in broilers.
