摘 要:在不同浓度缺铁水培条件下,采用电感耦合等离子体原子发射光谱仪(ICP-AES)测定转基因和非转基因植株叶片和根系Ca2+、Mg2+、Ni2+的含量。研究结果表明:在Fe2+浓度为1μmol.L-1时,转番茄铁载体蛋白基因(LeIRT2)八棱海棠株系叶片中Mg2+的含量显著低于非转基因株系,而根系Mg2+的含量显著高于非转基因株系;该浓度下转基因株系根部Ni2+含量显著高于非转基因株系,叶片中Ni2+含量与非转基因株系无显著差异。Fe2+浓度为10μmol.L-1时,转基因株系叶片和根系Mg2+、Ca2+的含量与非转基因株系无显著差异。Fe2+浓度为1μmol.L-1时,转基因株系叶片和根系Ca2+的含量与非转基因株系无显著差异。无铁条件下在叶片和根系中Ca2+、Mg2+含量无显著差异。The transgenic Malus robusta plants with LeIRT2 were selected for further study.The contents of Ca2+,Mg2+and Ni2+ in leaves and roots of the transgenic lines and non-transgenic plants were detected with the method of ICP-AES.The results showed that with the iron concentration of 1 μmol·L-1,the content of Mg2+ decreased sharply in leaves but increased notably in roots,compared with the non-transgenic plants.At the same concentration,the content of Ni2+ in roots was significantly higher than that of the compares,while with no obvious difference in leaves.There were no significant difference of the Mg2+ and Ca2+ content in leaves or roots between the transgenic and wild plants treated by iron with concentration of 10 μmol·L-1.There were no significant difference of the Ca2+ content in leaves or roots between the transgenic and wild plants treated by iron with concentration of 1 μmol·L-1.There were no significance differences of Ca2+,Mg2+ content in leaves or roots under iron-free conditions.
