摘  要:目的观察卵巢早衰(premature ovarian failure,POF)患者外周血中TH17细胞与FOXP3+CD25highCD4+调节性T(Treg)细胞比例及相关因子表达的变化并探讨其意义。方法取30例POF患者及12例健康妇女外周血,流式细胞术检测TH17和Treg细胞占CD4+T细胞的比例;实时定量PCR测定外周血单个核细胞(PBMC)中TH17和Treg细胞相关因子IL-17、RORγt、FOXP3、TGF-β、IL-1β及IL-6 mRNA的相对表达量。结果 POF患者外周血中TH17/CD4+T细胞的比例明显高于健康妇女(P<0.01),Treg/CD4+T细胞的比例较健康妇女有增高趋势但无统计学差异,TH17/Treg细胞比例的比值明显高于健康妇女(P<0.01)。POF患者PBMCs中,IL-17、RORγt、FOXP3的mRNA相对表达量明显高于健康妇女(P<0.01),而IL-1β、IL-6则明显低于健康妇女(P<0.01,P<0.05),TGF-β无明显差异(P>0.05)。结论 POF患者可能处于一种TH17细胞明显增多并趋于优势地位的炎性状态,这一机制可能参与了POF的发生、发展,但其中细胞因子的作用尚待进一步研究。Objective To observe the changes of TH17 and FOXP3+ CD25high CD4+ regulatory T cells(Treg) and TH17/Treg-related factors in peripheral blood of the patients with premature ovarian failure(POF),and explore the effects of peripheral TH17 and Treg cells in pathogenesis of POF.Methods The blood samples were collected from 30 POF patients and 12 healthy women.The ratios of TH17 and Treg cells in peripheral blood samples were examined with flow cytometry.The mRNA relative quantities of IL-17,RORγt,FOXP3,TGF-β,IL-1β and IL-6 were examined by using real-time PCR.Results Compared with healthy controls,the levels of TH17 cells in POF patients were much higher(P0.01),and the ratios of TH17/Treg increased significantly(P0.01).The mRNA relative quantities of IL-17,RORγt and FOXP3 in POF patients were much higher than those of healthy controls,while the mRNA relative quantity of TGF-β was similar with healthy controls(P0.05),and IL-1β and IL-6 were significantly lower than healthy controls(P0.01,P0.05).Conclusion In the POF patients TH17-dominant inflammatory status may involved in the pathogenesis of disease.
