摘  要:目的研究大鼠心肌缺血再灌注损伤后p38 MAPK的变化及纳洛酮对其的影响。方法 SD大鼠24只随机均分为假手术组(sham)、缺血再灌注组(IR)、纳洛酮组(Nal)。采用结扎左冠状动脉左前降支30 min,再灌注2 h制作心肌缺血再灌注模型。观察心电图J点电位的变化;Western Bloting检测p38 MAPK、p-p38 MAPK的表达;TUNEL法检测心肌细胞凋亡。结果 IR组大鼠的心电图J点电位明显抬高38 MAPK、p-p38 MAPK蛋白表达增强;细胞凋亡明显增多。Nal组大鼠的心电图J点电位抬高不明显38 MAPK、p-p38 MAPK蛋白的表达减弱;细胞凋亡明显减少。结论纳洛酮对大鼠心肌缺血再灌注损伤有保护作用,其机制可能与抑制p38 MAPK、p-p38 MAPK蛋白的表达及减少细胞凋亡有关。OBJECTIVE To investigate the change of p38 MAPK after myocardial ischemia reperfusion injury in rats and the effects of naloxone.METHODS Twenty-four SD rats were randomly divided into three groups.sham-operated group,IR group,IR and treated with Naloxone group.The myocardial ischemia reperfusion injury model was established by ligating the left anterior descending coronary artery for 30 minutes followed by reperfusion for 2 h.ECG was used to detect J dot changes.Terminal deoxynucleotidy transferase mediated dUTP fluorescein nick end labeling(TUNEL) and Western bloting were used respectively to detect the cardiomyocyte apoptosis and protein expressin of p38 MAPK and p-p38 MAPK,respectively.RESULTS In IR group ECG showed that J dot was significantly higher,and Western bloting sections showed that the expression of p38 MAPK and p-p38 MAPK were significantly higher,and TUNEL sections showed that the number of apoptotic cardiomyocytes was significantly increased,as compared to sham-operated group(P0.01).In Nal group ECG showed that J dot was significantly reduced,and Western bloting sections showed that the expression of p38 MAPK and p-p38 MAPK were significantly lower,and TUNEL sections showed that the number of apoptotic cardiomyocytes was significantly decreased,as compared to IR group(P0.01).CONCLUSION Naloxone has protective effect on cardiac muscle after the myocardial ischemia reperfusion injury,and its mechanism may be related to inhibition of the expression of p38 MAPK and p-p38 MAPK and reduction of cardiomyocyte apoptosis in rat myocardial ischemia reperfusion.
