摘  要:目的:了解台州市Yamagata系乙型流感病毒的基因特性、变异情况和WHO推荐流感疫苗株对人体的保护情况。方法:用狗肾传代细胞(MDCK)分离流感病毒,用标准血清及荧光定量RT-PCR方法鉴定。选2011年分离株进行HA1基因序列测定,用DNAman和DNAstar生物学软件进行序列分析。结果:2011年分离株HA1基因与参考毒株核苷酸同源性为88.6%~96.9%,氨基酸同源性为89.2%~98.0%,存在8个潜在的糖基化位点,比同谱系疫苗株在197位增加了一个糖基化位点"NKT"。与Yamagata系代表株在进化树的同一支上,与Victoria系代表株比,氨基酸162位缺失K。结论:本地2011年分离株与近年来WHO推荐的流感疫苗株在HA1片段有明显的氨基酸变异。2010年-2011年台州市主要分离到Yamagata系乙型流感病毒,而WHO推荐使用的2010年-2011年乙型流感疫苗株B/Brisbane/60/2008属于Victoria系,因此本年度流感疫苗对我市乙型流感不能提供最佳的保护性。Objective: To analyze the genetic characteristics and variation influenza B virus isolated in Taizhou and evaluate the protective effect of the vaccine strains recommended by WHO.Methods: Influenza viruses were isolated in MDCK cell and identified by standard serum and real-time RT-PCR. Isolates in 2011 were selected to conduct HA1 gene sequencing,DNAman and DNAstar were used to conduct sequence analysis.Results: The homology of nucleotide acids was 88.6%~96.9% between the isolated virus strains and reference strains,their amino acids homology was 89.2%~98.0%.HA1 gene of isolated strains had one more glycoaylation site than Yamagata-like vaccine strains at 197 site,and less K at 162 site compared to Victoria-like vaccine strains.Conclusion: Some antigenic drift occurred in HA1 gene of isolated strains.The protective effect of the vaccine strain was poor in 2010-2011 because the isolated strains and vaccine strains did not belong to the same lineage.
