摘 要:目的:探究福州市麻疹病毒基因的遗传变异特点及种系进化分布,为控制和消除麻疹提供科学依据。方法:应用Vero/slam细胞进行麻疹病毒分离,用RT-PCR方法扩增分离株病毒的N基因C末端450个核苷酸片段,扩增产物经纯化后,进行N基因核苷酸序列测定并推导出其aa序列,然后用DNASTAR MegAlign序列分析软件进行基因种系进化特性分析。结果:福州市麻疹分离株均为H1a基因亚型,3株病毒核苷酸同源性为99.3%~100%,在遗传树图中分属于1个独立分支,与疫苗株S191相比核苷酸同源性为87.5%,aa同源性为91.9%~92.1%。结论:福州麻疹野病毒与疫苗株S191在核苷酸与aa序列均存在较大差异,在麻疹监测工作中应进一步加强麻疹病原学的监测和麻疹遗传变异特性的研究。Objective: To understand the genetic variation characteristics of measles viruses and evolutionary distribution.Methods: Measles viruses were isolated with Vero/slam cell.The 450 C-terminal nucleotide fragment of the nucleoprotein(N) gene was amplified by RT-PCR and PCR products were purified and conducted determination of nucleotide sequences of N gene to deduce the amino acid sequences via bio-informatics.Finally the phylogenetic tree was analyzed with DNASTAR MegAlign soft ware.Results: The Measles viruses isolated in Fuzhou were identified as H1 genotype,H1a sub-genotype,which belong to one branch in the phylogenetic tree.The sequence identity matrix of the 3 isolates showed that the homology of nucleotide sequence was 99.3%~100.0%.The homology of nucleotide and amino acid sequences were 87.5% and 91.9%~92.1% respectively,compared with that of S191.Conclusion: The Measles viruses strains were obviously different from Shanghai 191 strain in nucleotide sequences and amino acid sequences.It should be taken for further monitoring and research on variation in measles viruses.
