摘 要:目的:提高发光菌急性毒性测试数据更具可靠性和可重复性。方法:以费舍尔弧菌(Vibrio fischeri)为受试生物的生物毒性测试质量控制方法,确定了菌液原始发光值和实验空白光损率对测定结果的影响以及阳性对照物控制范围。结果:为使测试结果有更高的重现性,菌液初始发光强度应大于500000;实验空白光损率应保持在50%±10%范围内;1.07 mg/L的Zn2+溶液阳性对照质控样应该控制在51%±11%范围内。结论:为使测试结果更加准确、稳定,应对菌液初始发光强度、实验空白光损率和Zn2+溶液阳性对照测试值加以控制。Objective: To make the luminescent bacteria toxicity test more reliable and repeatable.Methods:The quality control methods for luminescent bacteria toxicity test using Vibrio fischeri was studied,including the influences of original bacilli light-emitting and blank loss rate on the experimental results as well as the positive control of the control range.Results:In order to make the test more reproducible,the initial bacilli luminous intensity should be greater than 500000;the blank optical loss rate should be maintained in the range of 50%±10%;the result of the test for 1.07 mg/L of Zn2+ positive solution should be controlled in the range of 51%±11%.Conclusion:The initial luminous intensity,blank optical loss rate and Zn2+ positive solution should be under control while analyzing,making the reaults of luminescent bacteria toxicity test more accurate and stable.
