摘 要:目的:2010年杭州市发生了两起急性出血性结膜炎(Acute Hemorrhagic Conjunctivitis,AHC)疫情,为鉴定引起疫情的致病病原体。方法:采集了9份眼结膜拭子标本,使用实时荧光PCR或RT-PCR法分别检测临床标本中腺病毒、人肠道病毒70型(Human Enterovirus 70,HEV70)和柯萨奇病毒A组24型变种(CoxsackievirusA24 variant,CVA24v)的基因。使用Hep-2细胞和RD细胞分离病毒。对分离到的CVA24v毒株进行全长VP1区扩增和核苷酸序列测定和分析。结果:实时荧光PCR或RT-PCR法检测临床标本,其中有6份检测结果为CVA24v阳性,阳性率为66.7%,而腺病毒和HEV70检测结果均为阴性。使用Hep-2细胞共分离到5株病毒,通过分子定型均鉴定为CVA24v。进化树图显示,两起疫情是由两个病毒传播链引起的。结论:本研究成功地分离到5株CVA24v毒株,为今后杭州市AHC的防治及研究奠定了一定的基础。Objective: In 2010,two outbreaks of acute hemorrhagic conjunctivitis(AHC) occurred in Hangzhou.In order to identify the etiology of these outbreaks.Methods: 9 eye conjunctival swabs were collected from 9 outpatient patients,and detected for adenovirus,human enterovirus 70(HEV70) and Coxsackievirus A24 variant(CVA24v) genes by using Realtime one step RT-PCR or PCR methods.Viral isolates were obtained from 9 clinical specimens by using HEp-2 cells and RD cells.All CVA24v isolates were performed by VP1 sequencing.Results: 6 samples were positive for CVA24v by using Realtime one step RT-PCR or PCR methods.The positive rate was 66.7%,but none was positive for HEV70 and adenovirus.5 viral isolates were obtained from 9 clinical specimens by using HEp-2 cells,and all were identified as CVA24v by molecular typing method.Phylogenetic analysis of 5 CVA24v revealed that twice outbreaks were caused by at least 2 viral transmission chains.Conclusion: This study succeed in isolating 5 CVA24v viral,which was most important to prevent and control the outbreaks of AHC in Hangzhou.
