摘  要:目的:探讨吗啡(morphine,Mor)点燃条件位置性偏爱(conditioned place preference,CPP)重现大鼠海马CAl区N-甲基-D-天冬氨酸受体(NMDA受体)调节亚基NR2B,钙依赖钙调蛋白激Ⅱ(Ca^(2+)/calmodulin dependent protein kinase Ⅱ,CaMKⅡ)表达的变化。方法:用恒量法(10 mg·kg^(-1))给大鼠连续颈背部皮下注射(subcutaneous,sc)吗啡8 d建立CPP模型;用生理盐水替代吗啡训练大鼠10 d,使形成的CPP逐渐消退;单次sc2.5 mg·kg^(-1)吗啡点燃已消退的CPP。用免疫组化法检测吗啡点燃CPP重现大鼠海马CA1区NR2B、CaMKⅡ表达,图像分析系统测定阳性反应产物的平均灰度值的变化。结果:sc10 mg·kg^(-1)吗啡8 d建立CPP,生理盐水训练10 d使已形成的CPP消退,小剂量吗啡(2.5 mg·kg^(-1))使消退的CPP重现;吗啡点燃(CPP重现大鼠海马CA1区NA2B、CaMKⅡ表达,与对照组比较呈显著增加(P<0.05)。结论:小剂量吗啡诱发大鼠CPP重现行为可能与海马CA1区NR2B、CAMKⅡ表达增加有关。OBJECrIVE To study the changes of the protein expression of NR2B and CaMKⅡ in hippocampus CA1 area of rats with recurrence of conditioned place preference (CPP) induced by morphine. METHODS Morphine was administered via subcutaneous injection at constant dose (10 rag. kg-1 ) for 8 days to establish morphine CPP. The rats were administered saline instead of morphine to induce CPP extinction for 10 days. CPP was reinstated following a single priming injection of morphine (2. 5 mg.kg-1 ). The morphine induced rat CPP acquisition, extinction and reinstatement model was established. The protein levels of NR2B subunit and CaMK Ⅱ in hippocampus CA1 of rats with CPP recurrence of rats were measured by streptavidinperosidase immunohistochemical method and the mean gray values of immunoreactive product was detected by image analysis system. RESULTS Injecting morphine (10 mg.kg 1) for 8 days succeeded to induce CPP. After 10 days training with saline, CPP failed to be induced. A single priming injection of morphine(2. 5 mg/kg)kindled CPP. During CPP reinstatement, the expression of NR2B subunit and CaMK Ⅱ in hippocampus CA1 increased significantly compared with the saline group (P〈0. 05). CONCLUSION The increased expression of NR2B subunit and CaMK lI in hippocampus CA1 of rats involves in CPP recurrence induced by morphine.
