摘  要:铜转运蛋白(Copper transporter,COPT)属于CTR/COPT铜转运家族,参与调节生物体内铜的动态平衡。根据抑制性消减杂交文库分离的EST片段,采用3'-RACE技术从巴西橡胶树胶乳中获得HbCOPT5基因的全长cDNA序列,其ORF长420 bp,共编码139个氨基酸,预测HbCOPT5蛋白分子量为15.67 ku,等电点为5.26,含有3个跨膜区。进化树分析结果表明,HbCOPT5与蓖麻、杨树中的COPT蛋白亲缘关系最近;在拟南芥的6个COPT蛋白中,与AtCOPT5的亲缘关系最近。RT-PCR结果显示,健康植株中HbCOPT5的表达量是死皮树中的2.8倍。High-affinity copper transporter(COPT) is the key member of CTR/COPT family, which plays an important role in mediating copper homeostasis in organisms. Using the primers designed from the known EST from a suppression subtractive hybridization-cDNA library, the full-length cDNA sequence of HbCOPT5 was cloned from Hevea brasiliensis latex by 3-RACE technique. The opening reading frame(ORF)of HbCOPT5 was 420 bp in length, encoding 139 amino acids. HbCOPT5 was predicted to contain three transmembrane domains (TMDs) with molecular weight 15.67 ku and theoretical pI 5.26. Phylogenetic reconstruction revealed that HbCOPT5 was more closely related to the COPTs from Ricinu~ communis and Populus trichocarpa, and most closely related to the Arabidopsis COPT5 among the AtCOPT1-AtCOPT6 transporter family. In addition, HbCOPT5 exhibited 2.8 times higher transcriptional expression level in healthy trees than in tapping panel dryness trees.
