不同来源沙门菌入侵基因invH的检测及序列分析

摘  要:根据沙门菌invH基因序列设计1对引物,对9株沙门菌和4株常见病原菌进行PCR扩增,并将扩增出的in-vH基因进行克隆及序列分析,应用生物信息学方法预测invH蛋白的T细胞抗原表位。结果表明,9株沙门菌PCR均扩增出519bp的特异条带,4株非沙门菌均无特异带扩增;核苷酸序列分析证实,3株鼠伤寒沙门菌之间同源性为99.8%,与鸡白痢和鸡肠炎沙门菌之间的核苷酸序列同源性为98.6%,3株猪伤寒沙门菌和3株鸡源性沙门菌同源性较低,且不在同一支系统进化树上;invH蛋白有5个重复率最高的T淋巴细胞受体识别的抗原表位。A pair of oligonucleotide primer was synthesized according to the invH gene nucleotide sequence of Salmonella spp..And the invH gene was amplified from 9 strains of Salmonella and other 4 strains of pathogenic bacterium by PCR.Then the amplified invH gene was also cloned and sequenced.Based on bioinformatic techniuque,the antigenicity of T lymphcytes on invH protein was screened.The results showed the 519 bp specific bands was amplified from 9 strains of Salmonella by PCR,and no specific product from other 4 strains of pathogenic bacterium.The homology among three strains of S.typhinurium was 99.8%,among the three strains of S.typhinurium and Salmonella pollorum and Salmonella enteritidis was 98.6%.But the homology of the three S.typhisuis and three chicken-derived Salmonella was low,and the two derivations of Salmonella were not in the same phylogenetic tree.There were five higest repeated T cell epitope on invH protein.
