铜孵育对鸡肝脏线粒体呼吸功能的影响

摘  要:以CuSO4.5H2O为铜源,用含Cu2+终浓度为0μmol/L(A组)、5μmol/L(B组)、10μmol/L(C组)、15μmol/L(D组)、20μmol/L(E组)和30μmol/L(F组)的孵育介质对从肉鸡肝脏提取的线粒体进行孵育,每个铜浓度下孵育时间为10,25,40min;采用Clark氧电极法对孵育后的线粒体进行呼吸功能测定:呼吸控制率(RCR)、氧化磷酸化效率(OPR)和磷氧比(P/O)。结果显示,随着铜离子浓度的增加和孵育时间的延长,线粒体RCR,P/O和OPR这3项指标均出现不同程度降低;表明铜引起体外孵育的鸡肝脏线粒体功能损伤,且呈明显的时间、剂量效应。To further understand the biologic functions of copper in chichen mitochondria,copper sulfate anhydrous was used as the source of copper and mitochondrial respiration was observed after incubating the mitochondria which was freshly isolated from broiler hepatocyte with 0 μmol/L(Group A),5 μmol/L(Group B),10 μmol/L(Group C),15 μmol/L(Group D),20 μmol/L(Group E) and 30 μmol/L(Group F) of final concentration copper for 10,25 and 40 min.RCR,P/O and OPR were measured as index of mitochondrial respiration function by using Clark's method.The results showed that mitochondrial respiration function decreased dramatically with the increasing of copper concentration and incubating time.It is indicated that copper is harmful to the function of mitochondria and displayed a time-and dose affection.
