甘草附子汤对去卵巢大鼠骨组织中Ⅰ型胶原代谢及组织蛋白酶K表达的影响

摘　　要：目的研究甘草附子汤对绝经后骨质疏松的防治作用。方法 3月龄雌性大鼠随机分为假手术组、模型组、甘草附子汤高、低剂量组和雌激素组。造模成功后,假手术组和模型组给予等量水灌胃,甘草附子汤组给予甘草附子汤灌胃,雌激素组给予苯甲酸雌二醇肌注。3月后,测定大鼠Ⅰ型胶原(ColⅠ)、组织蛋白酶K(Cath K)、Ⅰ型前胶原羧基末端前肽(PICP)、Ⅰ型胶原交联羧基末端肽(ICTP)及骨小梁的变化。结果甘草附子汤组与雌激素组能显著增加大鼠的骨小梁的数量(P<0.05),降低Cath K的表达,且甘草附子汤高剂量组与雌激素组无明显差异(P>0.05),低剂量组不如高剂量组明显。结论甘草附子汤能够下调破骨细胞Cath K的表达来抑制骨基质中ColⅠ的分解,对骨质疏松症起到一定的治疗作用。Purpose To study the mechanism of Gancaofuzi Decoction on osteoporosis rats induced by ovariectomy.Methods 40 three-month-old female rats were divided into 5 groups：normal control group,ovariectomized group,estrogen group,low dose and high dose group of Gancaofuzi Decoction.After establishing model,the rats of Gancaofuzi Decoction group were given the intragastric administration with Gancaofuzi Decoction.The rats of estrogen group were given nilestriol,and other groups were given distilled water.After treatment for 3 months,the typeⅠcollagen（ColⅠ）,the cathepsinK（Cath K）,the content of carboxyterminal propeptide of typeⅠprocollagen（PICP）,carboxyterminal telopeptide of typeⅠcollagen（ICTP）and bone trabecula were measured.Results Compared with ovariectomized group,the bone trabecula of all over the body and ColⅠ in both Gancaofuzi Decoction group and estrogen group increased obviously（P〈0.05）.Moreover,compared with the ovariectomized group,Gancaofuzi Decoction can reduce the volume of ICTP,cathK,and increase the content of PICP inversely（P〈0.05）.Conclusion Gancaofuzi Decoction had the obvious treatment function to ovariectomy-induced osteoporosis,and its mechanism was to reduce Cath K level,which inhibited bone resorption and promoted bone formation.
