摘  要:目的采用响应面分析法优化白囊耙齿菌中腺苷的提取工艺。方法在单因素实验的基础上,以提取温度、提取时间、水料比为随机因子,腺苷得率为响应值,进行三因素三水平的Box-Behnken中心组合设计,采用响应面法分析三个因素对响应值的影响,并确定最佳提取工艺条件。结果白囊耙齿菌中腺苷的最佳提取工艺条件为:提取温度39.4℃,提取时间3.4 h,料水比50.4∶1,在此条件下腺苷得率的验证值为1.09%,与预测值1.11%之间的相对误差为1.80%。结论响应面法可较好地预测白囊耙齿菌中腺苷的提取工艺。Purpose Response surface methodology(RSM)was applied to optimize the extraction conditions of adenosine in Irpex lacteus mycelium.Methods Extracting temperature,extracting time and the solvent/solid ratio were selected as causal factors.The yield of adenosine was used as the responsive values.Based on the single factors experiments,a three-factor-three-level experiments design has been developed by Box-Benhken central composite design method.The response surface method was employed to analyze the results of experiments.Results The optimum extraction conditions of adenosine were as follows:extracting temperature was 39.4 ℃,extracting time was 3.4 h and solvent/solid ratio was 50.4∶1.The predictive maximum yield of adenosine was 1.11 %.The average yield of adenosine in 3 validation experiments was 1.09 %.The relative error was 1.80 %.Conclusion RSM analysis optimizing the extraction conditions for adenosine in Irpex lacteus mycelium is feasible.
