摘 要:为了研究闽桂地区尖凹大叶蝉不同地理种群的遗传分化程度,对闽桂地区尖凹大叶蝉5个不同地理种群的线粒体COⅡ基因部分片段进行了扩增、测序及序列分析。结果表明,获得的COⅡ基因590bp的序列中,碱基A,T,C,G平均含量分别为38.8%、33.8%、16.4%和11.1%,A+T含量明显高于G+C含量,表现出强烈的A、T偏向性,密码子第3位点的A+T含量高达87.6%。碱基替换主要发生在密码子第3位,转换大于颠换,且转换多发生在T-C间。尖凹大叶蝉群体内COⅡ基因具有丰富的遗传多态性,且各地理种群间已产生不同程度的遗传分化。中性检测显示尖凹大叶蝉进化遵循中性模型,且在过去呈现种群扩张趋势。尖凹大叶蝉不同地理种群之间的遗传距离在0.002~0.042。种群系统发育树显示出尖凹大叶蝉各地理种群的遗传距离符合所处的地理区划,与地理距离间不具有明显的相关性,推断遗传分化的产生主要取决于所处的生态环境。The partial fragments of mitochondrial COⅡ gene of five different Bothrogonia acuminata geographic populations were amplified,sequenced and analyzed to study genetic differentiation degree of different B.acuminata geographic populations in Fujian and Guangxi province.The results showed that the mean A,T,C and G content in 590 bp sequence of COⅡ gene was 38.8%,33.8%,16.4% and 11.1% respectively and A+T content was higher than G+C content obviously,the A+T content at the third locus of codon reached 87.6%,base substitution occurred at the third locus mainly,its transition was more than transversion and transition occurred between T and C mostly.The COⅡ gene in B.acuminata is of abundant genetic polymorphism and there is different genetic differentiation degree among different geographic populations.The evolution of B.acuminata followed the neutral model and there was an expansion trend of population.The genetic distance between different B.acuminata geographic populations is 0.002~0.042,which accords with its geographical division and there is no obvious correlation between geographical distances.Therefore,the genetic differentiation degree mainly depends on its ecological environment.
