益气解毒活络法对早期糖尿病肾病大鼠血清脂联素与血清超敏C-反应蛋白影响的实验研究

摘　　要：目的:通过观察益气解毒活络中药复方对早期糖尿病肾病(DN)大鼠血清超敏C-反应蛋白(hs-CRP)与血清脂联素(ADPN)表达的影响,以探讨益气解毒活络中药复方防治早期糖尿病肾病的效果及其作用机制。方法:将实验用大鼠分为正常对照组、模型空白组(高脂高糖饲料+STZ)、中药复方预防组、中药低剂量治疗组、中药高剂量治疗组及阳性西药对照组,采用高糖高脂饲料喂养结合一次尾静脉注射链脲佐菌素(STZ)的造模方法复制出糖尿病肾病模型,各药物组干预四周后处死取血,用ELISA方法测试各组血清hs-CRP与血清ADPN的表达情况,最后进行组间比较。结果:模型空白组大鼠的血清hs-CRP含量比正常组明显上升(P<0.01),其它各组经药物干预后,均可以显著降低其表达水平(P<0.01),但以中药低剂量治疗组效果最佳;模型空白组大鼠的血清ADPN比正常组明显下降(P<0.01),其它各组经药物干预后,均可以显著升高其表达水平(P<0.01),以中药复方预防组效果最佳。结论:DN的发病机制可能与血清hs-CRP和血清ADPN的表达异常密切相关;益气解毒活络中药复方防治DN发生发展的药理机制可能是通过调节血清hs-CRP和血清ADPN的异常表达来实现。Objective： To approach the effect and mechanism of treatment of invigorating vital energy,detoxicating and activating collaterals to the early period diabetic nephropathy by observing the expression of hypersensitive C reactive protein（Hs-CRP）and adiponectin（ADPN）in serum of diabetic nephropathy（DN） Rat.Methods： Divide the rats into 6 groups which are normal control,model blank group,prophylaxis group,LD group,HD group and positive control group.Build up the DN model by HG HF feeding and once injection through vena caudalis with STZ.Rat in every group accepted the action of every required interference factor for 4 weeks,and then executed to death and blood obtained.Measure the expression of ADNP and hs-CRP and then take the group comparison.Results： Expression of hs-CRP in model blank group is significantly higher than that in normal control（P0.01）,and result of the other groups effected by drugs shows signicantly lower than model blank group（P0.01）,especially in LD group.Expression of ADPN in model blank group is significantly lower than that in normal control（P0.01）,and result of the other groups effected by drugs shows signicantly higher than model blank group（P0.01）,especially in prophylaxis group.Conclusion： pathogenesy of DN may be closely related to the abnormal expression of hs-CRP and ADPN in serum.Prescription of invigorating vital energy,detoxicating and activating collaterals in prevent and cure to DN may by regulating the abnormal expression of hs-CRP and ADPN in serum.
