通络益肾温阳方对糖尿病肾病大鼠疗效的实验研究

摘　　要：目的:观察通络益肾温阳方对糖尿病肾病(DN)模型大鼠的疗效。方法:随机抽取9只大鼠作为正常对照组,行假手术;其余51只采用左侧单肾切除术合链脲佐菌素腹腔注射复制DN大鼠模型。将造模成功的47只大鼠分为病理对照组(16只)、中药治疗组(16只)、洛汀新+糖适平西药治疗组(15只),给药10 w。观察各组大鼠治疗前后一般情况,检测24 h尿蛋白定量、尿β2微球蛋白、空腹血糖、血清尿素氮、血肌酐;计算肾脏肥大指数,并行HE染色观察肾脏的病理组织学变化。结果:与对照组比较,中药治疗组大鼠一般状况明显改善,体重显著增加(P<0.01);检测24 h尿蛋白定量、尿β2微球蛋白、空腹血糖、血清尿素氮、血肌酐、肾脏肥大指数显著降低(P<0.01);肾病理组织学变化明显减轻。结论:通络益肾温阳方对DN模型大鼠具有一定的治疗效果。Objective：To observe therapeutic effect of Tongluo Yishen Wenyang recipe on diabetic nephropathy（DN）rat models.Methods：9 rats were selected as normal control group,51 rats were prepared DN models by left nephrectomy and peritoneal injection with streptozotocin.47 rats were divided into pathology control group（n=16）,TCM treatment group（n=16）and lotensin＋gliguidone treatment group（n=15）.10 w later,general states of rats were observed,24 h urine protein quantitate,contents of urinary ？2-microglobulin,blood-fasting sugar（BFS）,blood urea nitrogen（BUN）,serum creatinine（Scr）of rats were detected,renal hypertrophy index was calculated and renal changes of pathohistology were observed through HE staining.Results：Compared with pathology control group,general states and renal changes of pathohistology of rats were obviously improved,body weight of rats was increased（P0.01）,24 h urine protein quantitate,contents of urinary ？2-microglobulin,BFS,BUN and Scr,renal hypertrophy index of rats were decreased in TCM treatment group（P0.01）.Conclusion：Tongluo Yishen Wenyang recipe had positive therapeutic effect on DN model rats.
