摘 要:基于微流控芯片介电电泳(Dielectrophoresis,DEP)原理和技术,在自行设计制作的抛物线电极结构的微流控介电电泳芯片上,采用芯片介电泳临界频率测定法,选择缓冲液电导率为200~1000!S/cm,激发电压为5V,分别对红细胞(RBC)、白细胞(WBC)和死活HepG2肝癌细胞的临界频率进行了测试,检测相对误差小于5%(n=5),并对表征细胞生物状态的膜电导和膜电容进行了参数提取和分析。结果表明:不同种类的细胞及不同活性程度的细胞的细胞电学参数差异明显。利用本方法实现了对细胞种类及生物活性的定性判断和分析。On the basis of the principle and technology of microfluidic chip dielectrophoresis(DEP),crossover frequencies of red blood cell(RBC),white blood cell(WBC) and HepG2 were tested on a homemade parabolic electrode dielectrophoresis microfluidic chip when the conductivity of the buffers was in the range of 200-1000μS/cm and the applied excitation voltage at 5 V.Five parallel experiments were carried out and the relative error was less than 5%.Then,membrane conductance(Cmem) and membrane capacitance(Gmem) were calculated and analyzed.The result showed that electrical parameters of different types of cells and different levels of activity of cells were of great difference.In this study,the cell types and its biological activity was studied by using the cell DEP crossover frequency testing method.
