汉防己甲素滴眼液对眼组织毒副作用的实验研究

摘　　要：目的:探讨汉防己甲素(Tet)滴眼液对兔眼的毒副作用。方法:纯种新西兰白兔25只,雌雄各半,随机分为5组,每组5只。各组动物双眼结膜囊内分别滴入不同浓度(0.1%,0.5%,1%)的Tet滴眼液、赋形剂或生理盐水,每日4次,用药前及用药3月后对所有动物进行体重测量、眼前节裂隙灯显微镜和直接检眼镜检查以及闪光视网膜电图(ERG)检查,并观察动物用药后的进食、精神、活动情况,最后行组织学检查。结果:各组动物在用药期间进食、精神、活动未见明显改变;用药前后各组间体重差异无统计学意义(P>0.05)、直接检眼镜检查未见明显异常、闪光ERG检查中a、b波振幅及潜时的差异无统计学意义(P>0.05);组织切片检查未见异常;除1%Tet组的4只动物在用药初的3-4d出现Ⅰ级眼睑、结膜反应,Ⅰ级角膜反应外,其余均未见明显异常。结论:0.1%-1%Tet滴眼液对眼组织的毒副作用极小,临床应用安全。Objective：To probe the side effects of tetrandrine（Tet） eyedrop in rabbit eyes.Methods：A total of 25 rabbits（50 eyes） were randomly divided into 0.1%,0.5%,and 1% Tet groups,0.9% sodium chloride solution group and vehiculum group（10 eyes in each group）.The different eyedrops were respectively dropped into the double eyes of each rabbit four times daily for three months to examine the side effects of Tet eyedrop on rabbit eyes by ophthalmoscope,slit lamp microscope,electroretinograms（ERG）,and pathological sections.The weight of all animals were measured before treatment and three months after treatment.At the same time,the general condition（feed,mood,and activity） of all ainimals after treatment were observed.Results：The general condition of all animals didn＇t change significantly.No difference in the weight of animals,the a/b wave amplitude and latent period in the F-ERG were found among all the groups before and after treatment（P0.05）.There were no significant changes in pathological sections and the findings by direct ophthalmoscopy in all groups.Only four animals in 1% Tet group were observed gradeⅠreaction of corneal epithelium,conjunctival and eyelid.Conclusion：There were little side effects on rabbit eyes by using the 0.1%-1% Tet eyedrop.They can be safely used for clinical application.
