摘  要:应用大肠杆菌表达日本血吸虫抱雌沟蛋白(Schistosoma japonicum gynecophoral canal protein,SjGCP),将重组蛋白rSjGCP与FCA、ISA 206、ISA70M三种佐剂混合后免疫BALB/c小鼠,分析重组抗原诱导的免疫保护机制。用流式细胞术检测三次免疫后小鼠体内CD4+、CD8+T淋巴细胞亚群和IL-2、IL-4、γ-IFN、IL-10等细胞因子,应用酶联免疫吸附试验(ELISA)检测抗SjGCP特异性lgG抗体及其亚型lgG1水平。和空白对照组相比,GCP-FCA组和GCP-206组小鼠脾细胞中CD8+T淋巴细胞比例显著下降,而GCP-70M组的CD4+T淋巴细胞显著下降;GCP-FCA组、GCP-206组中IL-4、IL-10表达水平均显著上升;GCP-FCA组、GCP-206组、GCP-70M组血清中特异性IgG1亚型抗体含量显著升高。本研究为探讨重组日本血吸虫抱雌沟蛋白诱导的免疫保护机制积累了有价值的数据。For investigating the immune mechanism induced by the vaccine candidate Schistosoma japonicum gynecophoral canal protein(SjGCP), BALB/c mice were vaccinated with the recombinant protein SjGCP mixed with three different adjuvants FCA, ISA 206 and ISA 70M, respectively. The percentage of CD4+ and CD8+ T lymphocyte subsets and the cytokines IL-2, IL-4, γ-IFN and IL-10 in splenic lymphocytes from mice post three immunizations were analyzed by flow cytometry, and the serum of each groups were collected for detecting specific antibodies IgG and IgG1 by ELISA. The results showed that the percentage of CD4+ T cells in GCP-70M group and CDS+ T cells in GCP-FCA and GCP-206 groups decreased significantly post three immunizations, comparing to the PBS control group, while the percent of CD3+CD8-IL-4+ and CD3+CD8-IL-10+ cells in groups of GCP-FCA and GCP-206 increased. The ELISA results showed that the levels of the specific antibodies IgG 1 subtype in the sera of GCP-FCA, GCP-206 and GCP-70M groups were significantly higher than that of control groups. These results accumulated the valuable data for understanding the protection mechanism induced by recombinant protein SjGCP.
